Imazethapyr inhibition of acetolactate synthase inRhizobiumand its symbiosis with pea

Abstract: : Acetolactate synthase (ALS) activity extracted from Rhizobium leguminosarum biovar. viciae has been characterized. The optimum pH for extraction was 7É6 and for the assay 7É0. The for pyruvate was 7É2 mM, and the enzyme K m was saturated at 40 mM. An obligatory requirement of TPP and Mg2`for full ALS activity was observed. Valine was the only branched-chain amino acid that caused ALS feedback inhibition. The speciÐc activity of Rhizobium ALS was nearly 20 times the activity found in pea (Pisum sativum) leave… Show more

“…Furthermore, Leu, Ile, Val (the branched chain amino acids) increased in both cultivar nodules upon osmotic stress. ALS activity (the first enzyme in the branched chain amino acids pathway) in Rhizobium is high (Gonzalez et al 1996), when inhibitors of the enzyme (herbicides) are used and this contributes to a better survival of nitrogen fixing compared to nitrate-fed plants (Royuela et al 1998). Also, the aap double mutants of Rhizobium are unable to transport assimilated-N from bacteroids to the plant cells of the nodule.…”

“…Herbicide application can bring undesirable consequences to soil microorganisms, depending on the active compound, commercial formulation, dose (Royuela et al, 1998), climatic conditions and soil type (Silva et al, 2003). Data presented in Figure 1 show that number and dry weight of nodules/plant significantly increased with two hand hoeings and nonweeded treatments, compared to other treatments.…”

“…A aplicação de herbicidas em culturas que realizam simbiose com bactérias fixadoras de nitrogênio atmosférico pode prejudicar a eficiência na assimilação desse nutriente (Marenco et al, 1993;Royuela et al, 1998;Santos et al, 2004). Esses prejuízos devem-se à interferência do herbicida no metabolismo do microssimbionte, na planta hospedeira ou em ambos (Arruda et al, 2001).…”
