Imaging β-amyloid burden in aging and dementia

Abstract: Pittsburgh Compound B PET findings match histopathologic reports of beta-amyloid (Abeta) distribution in aging and dementia. Noninvasive longitudinal studies to better understand the role of amyloid deposition in the course of neurodegeneration and to determine if Abeta deposition in nondemented subjects is preclinical AD are now feasible. Our findings also suggest that Abeta may influence the development of dementia with Lewy bodies, and therefore strategies to reduce Abeta may benefit this condition.

“…PIB also demonstrated the ability to discriminate between healthy young controls and older patients, with no significant retention seen in controls [6]. Subsequent studies were able to demonstrate congruence of findings, with retention of PIB and diagnosis consistent with AD [7][8][9][10] . Equivocal findings in patients with PIB retention and clinical diagnoses of MCI are variable likely owing to cofounders, including amnesia, depression, and alternative dementias playing a role.…”

“…This increased retention is reported to be lower than that observed in AD subjects. The different increase in 11 C-PIB PET retention reported in normal controls, AD and DLB patients reflects a broad range of Aβ deposition [31] which may be related to the degree of microglia activation and/or CX3CR1 expression [30].…”

“…Preliminary studies report that cognitively normal elderly subjects show an increased 11 C-PIB retention in the prefrontal and posterior cingulate/precuneus regions [31]. This increased retention is reported to be lower than that observed in AD subjects.…”
