IL‐1α Counteract TGF‐β Regulated Genes and Pathways in Human Fibroblasts

Abstract: Dysregulated wound healing is commonly associated with excessive fibrosis. Connective tissue growth factor (CTGF/CCN2) is characteristically overexpressed in fibrotic diseases and stimulated by transforming growth factor-β (TGF-β) in dermal fibroblasts. We previously showed that interleukin-1 (IL-1α) counteracts TGF-β-stimulated CTGF mRNA and protein expression in these cells. The aim of this study was to explore the effects of IL-1α on further genes and pathways in TGF-β regulated fibroblasts. Transcriptional… Show more

“…Changes in cell activities and matrix components are usually associated with alteration in growth factor synthesis. TGF- β is an important tissue marker that can be produced by various cells in response to an injury and is typically produced for short periods and in limited amounts, since its action takes place in distinctive phases of the process [ 55 , 56 ]. Our results showed large amounts of TGF- β produced by groups receiving a high-fat diet and alcohol, contradicting the above statement that its production would be short and limited.…”

“…We believed that skin mast cells play a fundamental role in the regulation of inflammation due to the secretion of chemical mediators, mainly in the production of anti-inflammatory molecules that work directly on the resolution of inflammation by promoting cell proliferation, differentiation, migration and tissue remodeling [ 60 ]. TGF-β and IL-10 are considered universal mediators that can be synthesized by different cells, with positive action in the cutaneous repair process due to their capacity to accelerate matrix synthesis and tissue remodeling [ 61 , 62 ]. The results of our study corroborate this proliferative and remodeling effect, since the groups that received S pseudoquina extract showed increased amount of collagen and predominance of type I fibers, which give the tissue strength and resistance.…”

“…The increase in cellularity and in mast cells number can justify the increase in TGF- β expression that was observed in this study [ 15 ]. TGF- β is considered to be a universal mediator that can be synthesized by different cells with positive action in the cutaneous repair process due its anti-inflammatory effect and capacity to decrease cell migration and accelerate the matrix synthesis [ 24 , 71 ].…”
