IgG‐Type Cold Agglutinins in Children and Corresponding Antigens

Abstract: Abstract. Two IgG‐type cold auto‐agglutinins causing transiently occurring haemolytic anaemia in children are described. The antibodies were limited to the IgG fraction obtained by DEAE cellulose chromatography and were resistant to 2‐mercaptoethanol treatment. One auto‐agglutinin reacted ‘incompletely’ (in the enzyme test), the other reacted ‘completely’ and its reactivity was furthermore abolished by treatment of red cells with proteolytic enzymes, but not with neuraminidase. Because of the inactivation of … Show more

“…In one patient (JSh), the serolo gy was unusual in that the IgG autoanti body reacted much better at room temper ature than at 37 °C; a few other examples of IgG cold autoantibodies causing hae molysis in children have been noted [29,30],…”

“…Details of this case are being prepared for publication (Lockyer and Webb). An exceptional hightitre cold agglutinin which as found to have a new specificity anti-Pra by Roelke et al [8] was confined to the IgG fraction, behaving similary to one of the enzyme auto-antibodies tested. Two anti-i cases had activity in both IgG and IgM fractions in agreement with Capra et al [2], Both were macro-globulinaemic, precipitated heavily in distilled water and caused abnormal shrinkage of the gel.…”

“…IgG antibodies can cause agglutination in saline medium, for example anti-D and anti-Kell [6,7] and anti-Pra [8], We had thought that the saline reaction of D--/D--cells with some incomplete anti-D sera was brought about by IgG antibody due to a reduced zeta-potential caused by cell membrane abnormalities. It was therefore surprising to find that the specific saline activity eluted with the IgM and IgA molecules from the gel.…”

“…The purified material strongly inhibited anti-P, antisera (Table 1) but did not inhibit the agglutination of normal human red cells by antisera of the specificities : anti-A, anti-B, anti-H, anti-I (6 sera examined), anti-i (3 sera examined), anti-P (2 sera examined), anti-Pr,, anti-Pr,, anti-Pr, [21]. (Table 4).…”
