Identification of α-1L Adrenoceptor in Rabbit Ear Artery

Abstract: The ␣-1L adrenoceptor (AR) was identified in rabbit ear artery by both functional and ligand binding studies. In functional studies using arterial rings, the contractile response to NS-49 [(R)-(Ϫ)-3Ј-(2-amino-1-hydroxyethyl)-4Ј-fluoromethanesulfonanilide hydrochloride] (␣-1A and ␣-1L AR-selective agonist) was competitively antagonized with low affinities by prazosin, RS-17053 [N-[2-(2-cyclopropylmethoxyphenoxy) ethyl]-5-chloro-␣,␣-dimethyl-1H-indole-3-ethamine hydrochloride], and 5-methylurapidil but with hi… Show more

“…1A), it is not likely that the high density estimated in the present study is an overestimation due to nonspecific accumulation of the radioligand into intact tissues and cells. Rather, underestimation may be caused by the conventional binding method with membrane preparations, as reported previously in the rat heart (16) and rabbit ear artery (21). We are now examining this discrepancy between different binding assay methods.…”

“…In functional studies, antagonist affinity estimates (pK B values) were determined for a single concentration of antagonist by the concentration ratio method, as reported previously (21). We used a one-way analysis of variance (ANOVA), followed by Scheffe's post-hoc test, or Student's paired or unpaired t-test where appropriate.…”

“…Rather, as described above, a close relationship between a1L-adrenoceptor and a1A-adrenoceptor has been considered (Ford et al, 1997;Hiraizumi-Hiraoka et al, 2004;Morishima et al, 2008).…”

“…However, tissue homogenization may cause changes in the receptor environment, resulting in measured properties that differ from those of the receptor in the native tissue. Recently, a binding assay that uses intact tissue segments was developed and was shown to be a powerful method for analysis of the inherent properties of receptors present in native tissues (7,15). In the intact segment binding assay, in contrast to conventional membrane binding assays, it is not necessary to consider either the yield loss of receptors or modification of the natural environment of receptors that may be caused upon tissue homogenization.…”
