Identification of vasculature‐specific genes by microarray analysis of etsrp/etv2 overexpressing zebrafish embryos

Abstract: We performed sequence homology and expression analysis for up-regulated genes that were novel or previously unassociated with the zebrafish vasculature formation. Angiotensin II type 2 receptor (agtr2), src homology 2 domain containing E (she), mannose receptor C1 (mrc1), endothelial cell-specific adhesion molecule (esam), yes-related kinase (yrk/fyn), zinc finger protein, multitype 2b (zfpm2b/fog2b), and stabilin 2 (stab2) were specifically expressed in vascular endothelial cells during early development whil… Show more

“…The zebrafish genome encodes two paralogs of Esam, esama and esamb. In zebrafish, esama expression has been reported in ECs, the endocardium and also in a subset of neurons in the brain, including the epiphysis/pineal gland (Wong et al, 2009). We confirmed the endothelium-specific expression of esama, whereas we observed only weak expression of esamb in the vasculature (Fig.…”

“…A second receptor [the ANG II type 2 (AT 2 ) receptor], proposed to modulate the actions of the AT 1 receptor, is found in abundance in early fetal development, but expression falls after birth. Similarly, expression of the zebrafish AT 2 receptor (agtr2), seen principally in the DA at 24 hpf, begins to diminish by 36 hpf (78). As expression of renin continues to increase after 36 hpf, the principal roles of renin/RAS in developing zebrafish, be it for ion homeostasis or other functions, may be via the AT 1 rather than AT 2 receptor.…”

“…Their model fits well with our current results in zebrafish. It is well known that Etv2 overexpression also results in endothelialization in zebrafish (35,37,42,59) and Xenopus (57). All of these observations suggest that Etv2 may play a conserved role in maintaining the endothelial characteristics of hemangioblasts in the early developmental periods.…”

“…S5B). Because etv2 has been shown to regulate the expression of endothelial genes positively (35)(36)(37), the expression of fli1a and other endothelial markers (flk1 and tie1) was analyzed at later developmental stages ( Fig. 3 B and C and Fig.…”
