Identification of the long non-coding RNA POU3F3 in plasma as a novel biomarker for diagnosis of esophageal squamous cell carcinoma

Abstract: BackgroundRecent studies have demonstrated that long non-coding RNAs (lncRNAs) were present in the blood of cancer patients and have shown great potential as powerful and non-invasive tumor markers. However, little is known about the value of lncRNAs in the diagnosis of esophageal squamous cell carcinoma (ESCC). We hypothesized that ESCC-related lncRNAs might be released into the circulation during tumor initiation and could be utilized to detect and monitor ESCC.MethodsTen lncRNAs (HOTAIR, AFAP1-AS1, POU3F3, … Show more

“…Emerging evidence has indicated that aberrant expression of lncRNAs is associated with numerous diseases, including diabetes (7,8). Importantly, lncRNAs were shown to act as regulators of tumorigenesis via participating in biologic processes, such as modulation of apoptosis, proliferation, drug resistance, and the epithelial-mesenchymal transition process (9)(10)(11)(12). For example, our previous study demonstrated that the lncRNA Hox transcript antisense intergenic RNA (HOTAIR) contributes to cisplatin resistance of human lung adenocarcinoma cells via downregulation of p21 WAF1/CIP1 expression.…”

“…Recently, several studies have demonstrated that lncRNAs are stably presented in the blood of cancer patients, such as lncRNA POU3F3 (28) and lncRNA TapSaki (29). Appearance of circulating lncRNA highlights the potential for noninvasive biomarkers derived from lncRNA expression profiles for prognosis and diagnosis of cancer, as these biomarkers have high sensitivity and specificity.…”

“…Later, Zhou et al reported the potential diagnostic value of plasma lncRNA H19 in gastric cancer [5]. In addition, in the field of esophageal squamous cell carcinoma diagnosis, plasma lncRNA POU3F3 was able to serve as a potential biomarker [12]. RNA fragments were shown to be released spontaneously from tumor cells; in vivo, a xenograft model validated that lncRNA fragments actively enter the circulatory system in mice.…”
