Identification of potential genes that contributed to the variation in the taxoid contents between two Taxus species (Taxus media and Taxus mairei)

Abstract: Taxol is currently a valuable anticancer drug; however, the accumulated mixture of taxoids can vary greatly among Taxus species. So far, there is very little genomic information for the genus Taxus, except for Taxus baccata. Transcriptome analysis is a powerful approach to explore the different regulatory mechanisms underlying the taxoid biosynthesis pathway in Taxus species. First, we quantified the variation in the taxoid contents between Taxus media and Taxus mairei. The contents of paclitaxel and 10-deacet… Show more

“…Therefore, the development of efficient and simultaneously green‐friendly approaches to reduce metal risks and toxicity in plants is a matter of special interest. Plant growth regulators control various important biological processes at physiological and molecular level (Peleg and Blumwald , Xu et al , Yu et al , Kumar et al , b). Nitric oxide (NO) is regarded as an important signaling compound, inducing a variety of developmental and physiological responses in plants, ranging from germination and senescence, to photosynthesis and redox homeostasis (Nabi et al ).…”

“…Fresh twigs of each sample were collected from three Taxus species, dried at 40°C for 3 d, and powdered. A modified version of a previously published method was used to prepare crude extracts [ 32]. In brief, 2.0 g powder of each sample was mixed with 30 mL of 100% methanol, and the mixture was subjected to ultrasonication for 60 min.…”

“…Other detailed parameters of the HPLC-MS/MS analysis were as follows: the capillary temperature was 270°C; the ion spray voltage was 3,000 V; the auxiliary gas and sheath gas was N 2 ; and the collision gas was high purity argon. Additionally, the positive electrospray ionization mode was employed and multiple-reaction monitoring was applied for the The separation and determination of eight flavonoids, including quercetin, luteolin, kaempferol, amentoflavone and ginkgetin, were performed according to the same UPLC-MS/MS method described in our paper [ 32].…”

“…Next-generation sequencing is a cost-efficient way to analyze transcriptomic variations under different treatment conditions (Shen et al, 2017;Yu et al, 2017). In 2012, expression profiling of P. cubensis-infected cucumber identified differential expressed genes (DEGs) during the infection process from 1 to 8 d post inoculation (Adhikari et al, 2012).…”
