Identification of orcokinins in single neurons in the stomatogastric nervous system of the crayfish, Cherax destructor

Abstract: The orcokinins are a highly conserved family of crustacean peptides that enhance hindgut contractions in the crayfish Orconectes limosus (Stangier et al. [1992] Peptides 13:859-864). By combining immunocytochemical and mass spectrometrical analysis of the stomatogastric nervous system (STNS) in the crayfish Cherax destructor, we show that multiple orcokinins are synthesized in single neurons. Immunocytochemistry demonstrated orcokinin-like immunoreactivity in all four ganglia of the STNS and in the pericardial… Show more

“…This is reminiscent of work on some other modulators that are also present in STG somata in young animals (FĂ©nelon et al, 1998Kilman et al, 1999). There are now a number of reports of STG somata stained for one or another neuromodulator Skiebe, 1999;Li et al, 2002;Skiebe et al, 2002). In these studies, as in this paper, STG somatic staining is relatively weak compared with either neuropil staining or staining of somata elsewhere in the nervous system.…”

“…However, in the crustacean stomatogastric system, no growth factors are presently known, and only one peptidergic neuromodulator has been shown to be produced by neurons within the STG. However, this modulator is not produced by neurons of the pyloric network itself (Skiebe et al 2002), and it is thus unlikely to act as "activity messenger" in the system.…”

“…This antibody has been used previously to map the distribution of orcokinins in the nervous system of numerous crustacean species (Bungart et al, 1994;Dircksen et al, 2000;Li et al, 2002;Pulver and Marder, 2002;Skiebe et al, 2002Skiebe et al, , 2003. To confirm its specificity for orcokininrelated peptides in C. productus, we preincubtaed the antibody with 10 -6 M orcokinin (Bachem; catalog No.…”
