Identification of novel surfactin derivatives from NRPS modification of Bacillus subtilis and its antifungal activity against Fusarium moniliforme

Abstract: BackgroundBacillus subtilis strain PB2-L1 produces the lipopeptide surfactin, a highly potent biosurfactant synthesized by a large multimodular nonribosomal peptide synthetase (NRPS). In the present study, the modules SrfA-A-Leu, SrfA-B-Asp, and SrfA-B-Leu from surfactin NRPS in B. subtilis BP2-L1 were successfully knocked-out using a temperature-sensitive plasmid, pKS2-mediated-based, homologous, recombination method.ResultsThree novel surfactin products were produced, individually lacking amino acid Leu-3, A… Show more

“…SPE70, SPE80, SPE90 and SPE100: fraction eluted with 70, 80, 90 and 100% methanol from SPE C 18 and the same letter means do not differ statistically (P < 0Á05) by Duncan tests. strong antifungal activity against Fusarium graminearum (Gong et al 2015), Botryosphaeria dothidea (Li et al 2016) and Botrytis cinerea (Zhang et al 2013), among others; surfactin analogues demonstrate high levels of suppressive activity against Fusarium oxysporum (Gu et al 2017), Fusarium moniliforme (Jiang et al 2016;Sarwar et al 2018), R. solanacearum (Xiong et al 2015), among others. In this study, the LP mixture (iturin, fengycin and surfactin) yielded by B. amyloliquefaciens strain FJAT-2349 could effectively inhibit the growth of R. solanacearum.…”

“…The culture was centrifuged (10,000 × g, 4 °C) for 10 min to remove LC. Then, 6 N HCL was added to adjust the pH of the supernatant (pH=2.0) and induce precipitation(42). Finally, the CLPs contained in the precipitation were fully dissolved in methanol, and a rotary evaporator (RE52CS-1, YARONG, Shanghai, China) was then used at 50°C and 65 rpm to obtain CLE for further analysis.…”

“…corresponding to C [14][15][16] Bacillomycin L, surfactin A and fengycin A were both proved to have the strong antifungal activity against a wide variety of fungi, especially for filamentous fungi (Ongena and Jacques 2008;Falardeau et al 2013;Jiang et al 2016). The three antifungal metabolites from LHSB1 could be responsible for its antifungal activity against S. rolfsii.…”
