Identification of MicroRNAs Dysregulated in CD14 Gene Silencing RAW264.7 Macrophage Cells

Abstract: A cluster of differentiation antigen 14 (CD14) is involved in lipopolysaccharide (LPS)-induced proinflammatory cytokine release and LPS-induced septic shock. MicroRNAs (miRNAs) are short non-coding RNAs that are involved in the epigenetic regulation of cellular process and bacterial infection. Our previous study indicated that siRNA against CD14 effectively inhibited LPS-induced tumor necrosis factor alpha, chemokine (C-X-C motif) ligand 2, interleukin-6 release, and NO production. To identify miRNAs which are… Show more

“…In CD14 + neonatal monocytes, enforced expression of miR-125b suppresses translation of TNF-α [67]. Silencing of CD14 by small interfering (si)RNA abolishes the production of TNF-α and IL-6 [68]. …”

“…Among them, elevated hsa-miR-199-5p was found to be involved in the progression of neoplastic lesions such as lung cancer and oral squamous cell carcinoma [29,30]. Furthermore, in a mouse model of sepsis, mmu-miR-199-5p correlated with the in ammatory response process of macrophages in innate immunity and surfactant protein D and exacerbates intestinal barrier dysfunction by inhibiting the NF-κB pathway [31,32]. These ndings suggest that elevation of hsa-miR-199-5p may also be associated with in ammation in sepsis.…”

“…Regardless, these mediators are similar and involved in many parts of the ACS cellular pathological mechanism. For example, CD14 gene silencing was reported to significantly change the expression of 199a-3p, miR-199a-5p, and miR-21-5p in the RAW264.7 macrophage cell line [ 91 ]. Endothelin-1 (ET-1) is a peptide encoded by the ET-1 gene (EDN1).…”
