Identification of Acremonium acutatum and Trichothecium roseum isolated from Grape with White Stain Symptom in Korea

Abstract: During 2010 and 2012 grape harvest seasons in Gyeonggi-do, Korea, a white stain symptom was observed on the harvested grape fruits in 'Campbell-Early' and 'Kyoho' varieties. In samples collected from the infected vine, two different strains of pathogenic fungi have been found and identified as Acremonium acutatum and Trichothecium roseum based on fungal morphology and nucleotide sequence of internal transcribed spacer (ITS) and supported by the phylogenetic analysis of the rDNA-ITS region. The DNA homologies o… Show more

“…Acremonium spp. are common soil and plant saprophytes [ 24 ], but some species are known as opportunistic human and animal pathogens [ 25 ] or as plant pathogens [ 26 ]. The species of this genus are morphologically characterized by colonies with slow or moderate growth; thin hyphae; simple or poorly basitonously branched, orthotropic phialides, usually gradually tapering toward the tip; and conidia that are usually small, unicellular, and aggregated in slimy heads, chains, or both [ 24 27 ].…”

“…Most enriched GO terms of up DEGs were single-organism process (315, 20.78%), single-organism cellular process (290, 19.13%), single-organism metabolic process (219, 14.45%), and catabolic process (102, 6.73%) in the biological process category. Additionally, genes involved in oxacid metabolism (70,4.62%) and oxi-reduction process (67, 4.42%) were also up regulated. High enrichment in the subcellular components localized in the cell (342, 22.56%), cell part (341, 22.49%), intracellular (320, 21.11%), and intracellular part (315, 2.078%).…”

“…Postharvest fungal diseases are responsible for quality deterioration of fruits and vegetables and for resultant severe losses of produce during handling, transportation, and storage. Phytopathogen Trichothecium roseum is one of the most significant spoilage fungi in China, causing decay in stored apples [1], melons [2,3], grapes [4], strawberries [5], and oranges [6]. Presently the pathogen can be controlled predominantly by chemical fungicides [7].…”
