Identification of differentially expressed genes in liver of marine medaka fish exposed to benzo[a]pyrene

Abstract: The construction of a subtractive cDNA library of organisms and the analysis thereof have proven meaningful in the search for clones whose expressions are regulated by exterior changes. The principal objective of this study was to identify the differentially expressed genes of marine medaka fish (Oryzias javanicus) under benzo[a]pyrene exposure conditions. Medaka fish were exposed to 100 μg/L of benzo[a]pyrene for 24 hr and its hepatic RNA was extracted; RNA from the livers of non-exposed medaka fish was also … Show more

“…These results were produced when Cd acted as a toxic stressor, and expres- sion of sHSPs was induced to maintain homeostasis and to protect cells. B [a]P has been shown to change the expression levels of HSP transcripts in the liver of medaka Oryzias javanicus fish (Won et al 2011). In this study, the expression level of Mm-HSP20 mRNA was significantly up-regulated in response to B [a]P in the digestive gland and hemocytes.…”

“…Based on the simultaneous suppression of amplification and the low amounts of RNA obtained from less abundant tissues, SSH has been proven to be useful in targeted screening of rare transcripts to find differentially expressed candidate genes in specific tissues (Rajput et al, 2013). In recent years, SSH has been successfully used in studies of infectious diseases, immune tolerance, metabolic regulation, and screening of genes related to the toxicity and stress of environmental pollutants (Dai et al, 2010;Liu and Sung, 2011;Won et al, 2011;Valdiglesias et al, 2012;Li et al, 2015). In this study, approximately 259 genes (169 known, 103 uncharacterized, and 34 unknown ESTs) were identified by constructing subtractive cDNA and enriched cDNA libraries from hepatic tissue after exposing fish to [C 16 mim]Cl.…”

“…The β-actin gene of Javanese medaka ( actb ; GenBank accession no. DQ660327) was used as an internal control following confirmation of stable transcription from the BPA exposure microarray data [ 38 ]. The group means were compared using ANOVA, followed by Duncan’s test for multiple comparisons.…”
