ABSTRACT--Morphologyand immunological roles of the three distinct types of hemocytes, hyaline, small granular and large granular cells in black tiger shrimp Penaeus monodon were stud ied. The study of functions of these hemocytes in the elimination of injected yeast (Saccharomy ces cerevisiae) or Vibrio harveyi as foreign bodies showed a rapid response against those particles. Together with fixed phagocytes, the blood cells removed the yeast through the process of phagocy tosis, nodule formation and encapsulation, which occurred at almost all parts of the body. The injection of the yeast caused a marked reduction in the blood cell counts in the hemolymph during the first 1 h. V. harveyi was efficiently removed within 3 h after injection. The granular cells (small granular and large granular hemocytes) were the major blood cells that are associated with phenoloxidase activity. The role of hemocytes and fixed phagocytes on defense mechanism in the shrimp were discussed.

We report here the isolation of three members of a new family of antimicrobial peptides from the hemolymph of shrimps Penaeus vannamei in which immune response has not been experimentally induced. The three molecules display antimicrobial activity against fungi and bacteria with a predominant activity against Gram-positive bacteria. The complete sequences of these peptides were determined by a combination of enzymatic cleavages, Edman degradation, mass spectrometry, and cDNA cloning using a hemocyte cDNA library. The mature molecules (50 and 62 residues) are characterized by an NH 2 -terminal domain rich in proline residues and a COOH-terminal domain containing three intramolecular disulfide bridges. One of these molecules is post-translationally modified by a pyroglutamic acid at the first position. Comparison of the data obtained from the cDNA clones and mass spectrometry showed that two of these peptides are probably COOHterminally amidated by elimination of a glycine residue. These molecules with no evident homology to other hitherto described antimicrobial peptides were named penaeidins.

Extreme events associated with global change will impose increasing stress on coastal organisms. How strong biological interactions such as the host-parasite arms-race are modulated by environmental change is largely unknown. The immune system of invertebrates, in particular phagocytosis and phenoloxidase activity response are key defence mechanisms against parasites, yet they may be sensitive to environmental perturbations. We here simulated an extreme event that mimicked the European heat wave in 2003 to investigate the eVect of environmental change on the immunocompetence of the mesograzer Idotea baltica. Unlike earlier studies, our experiment aimed at simulation of the natural situation as closely as possible by using long acclimation, a slow increase in temperature and a natural community setting including the animals' providence with natural food sources (Zostera marina and Fucus vesiculosus). Our results demonstrate that a simulated heat wave results in decreased immunocompetence of the mesograzer Idotea baltica, in particular a drop of phagocytosis by 50%. This suggests that global change has the potential to signiWcantly aVect host-parasite interactions.
