Identification of bla KPC-2 on different plasmids of three Morganella morganii isolates

Abstract: Three Morganella morganii strains resistant to carbapenems were recovered from the surgical intensive care unit (SICU) in our hospital. Carbapenemases and extended-spectrum β-lactamases (ESBLs) were respectively detected by the modified Hodge test and the modified Clinical and Laboratory Standards Institute (CLSI) ESBL confirmatory test in all isolates. Amplification of whole-cell and plasmid DNAs extracted from isolates with primers specific for the bla (KPC) produced an amplicon confi… Show more

“…Genes from M. morganii that encode cefotaxime-hydrolysing β-lactamases have also been reported with increasing frequency [42]. Plasmid-borne drug resistance factors have also increased the virulence of M. morganii [26,27]. Gene products that confer multidrug resistance, including metallo-β-lactamases and efflux pumps for tetracycline, tellurite, bicyclomycin, and kasugamycin, have been commonly reported for clinical isolates of M. morganii .…”

“…The drug resistance of M . morganii was introduced via extra genetic elements [26,27] and/or mobile elements [23,24]. The resistant strains that carry bla CTX-M gene are capable of producing β-lactamases [28], which can break down the extended spectrum β-lactam drugs [29].…”

“…Rifampicin-resistant Escherichia coli EC600 (LacZ − Nal r Rif r ) was used as the recipient strain. A conjugation experiment was carried out in mixed-broth cultures as described previously (Shi et al , 2012). Escherichia coli transconjugants were selected on Mueller–Hinton agar plates containing 256 µg rifampicin ml −1 (Sigma) plus 1 µg imipenem ml −1 .…”

“…Soon afterwards, these plasmid-mediated carbapenemases were identified in multiple genera and species of the Enterobacteriaceae and non-fermenters. The geographical distribution of bla KPC -producing isolates has spread not only within the USA (Bradford et al , 2004; Bratu et al , 2005), but also in Israel (Navon-Venezia et al , 2006), France (Naas et al , 2005), Greece (Cuzon et al , 2008), China (Shi et al , 2012), Argentina and Brazil (Monteiro et al , 2009).…”
