The pursuit of bioactive compounds from natural sources for potential pharmaceutical and therapeutic applications remains a prominent focus of scientific investigation. Among the myriad of botanical treasures, M. calabura, colloquially known as the Jamaican cherry or strawberry tree, stands as an intriguing candidate, owing to its extensive historical utilization in folk medicine across diverse cultures. This study aimed to comprehensively characterize the bioactive compound profile of M. calabura leaves by employing a spectrum of solvents with varying polarities for extraction. The choice of solvent during extraction plays a pivotal role in determining both the types and quantities of compounds obtained from plant material. Our methodology encompassed cutting-edge analytical techniques, notably gas chromatography-mass spectrometry (GC-MS), to unravel the intricate chemical composition of the extracts derived from M. calabura leaves. Moreover, we scrutinized the potential influence of solvent polarity on extraction efficiency and subsequent bioactivity of the isolated compounds. Our findings underscore the profound impact of solvent selection on the repertoire of bioactive compounds extracted from M. calabura leaves. Polar solvents exhibited a proclivity for extracting compounds of higher polarity, while non-polar solvents favored the extraction of less polar constituents. The identification of prominent bioactive compounds within these extracts encompassed delta-tocopherol, 7-(γ,γ-dimethylallyloxy)flavanone, Wogonin, Neophytadiene, Octadecanoic acid, and Hexatriacontane, each with its own distinctive pharmacological significance. This study thus illuminates the critical role of solvent polarity in optimizing extraction processes and maximizing the therapeutic potential of M. calabura. The insights gleaned from this investigation not only contribute to our understanding of natural product chemistry but also hold the promise of inspiring the development of novel pharmaceutical agents and therapeutic interventions with wide-ranging applications.

This study aims to analyze the phytochemical and antimicrobial-related assays including antibacterial, antifungal, antipellicle, and antibiofilm potential of the root of an alpine medicinal plant (Arnebia euchroma (Royle) I.M.Johnst). Ethanolic, methyl acetate, chloroform, and aqueous extracts of root parts of the A. euchroma plant were prepared. The active metabolites of the root part of the plant were evaluated both qualitatively and quantitatively by GC-MS and HPLC techniques, using ethanolic extract. The antimicrobial potential of A. euchroma was investigated using crude extract, fractions, and AgNPs of the A. euchroma root against two Gram-positive bacteria (Enterococcus, Staphylococcus aureus) four Gram-negative bacteria (Salmonella typhi, Pseudomonas aeruginosa, Escherichia coli, Proteus vulgaris) and five fungal strains (Rhizoctonia, Cuneate fasciculus, Aspergillus niger, Fusarium oxysporum, and Candida auris). The medicinal traits of Callicarpa macrophylla Vahl. have been used in traditional and folklore medicinal system, however, scientific evaluation of such medicinal plants have not been much explored yet. Different solvent extracts of the whole plant of C. macrophylla from the Himalayan region are the key subject of interest to evaluate and exhibit their antioxidant properties concerning its phytochemical composition. The different polarity extracts of C. macrophylla were prepared using the Soxhlet apparatus and both of their GC-MS profile outlined their individual components. The antioxidant activity of both extracts was evaluated for DPPH radical scavenging activity, metal chelating activity, and reducing power activity. Thirty-two compounds were identified in the hexane extract, accounting for 63.56% of the total extract composition with tetratetracontane (9.78%), as the prominent compound. A sum of 26 compounds making up 79.74% of the total methanolic extract composition was identified and the major compound was a diterpenoid, trachylobane (31.87%). The methanolic extract exerted potent antioxidant properties when compared, in vitro, to hexane extract as it showed better IC50 values of DPPH radical scavenging activity (91.97 ± 0.33 µg/ml), metal chelating activity (10.90 ± 1.40 µg/ml) and reducing power activity (56.72 ± 0.67 µg/ml). The antioxidant potential of the methanolic extract was also comparable to standards. Therefore, the methanolic extract exhibited better results as compared to hexane extract and had comparable potential with standards in vitro, if further explored might be utilized as an antioxidant agent in various pharmaceutical and cosmetic industries.
