Identification of Bio-oil Compound Utilizing Yeasts Through Phenotypic Microarray Screening

Abstract: Biomass pyrolysis bio-oil contains a plethora of carbon sources with the potential to be utilized by microorganisms and converted into high value products. However, the majority of these compounds are either highly toxic to microorganisms or are not directly utilizable. Hence research is required to develop methods of separating microbe friendly compounds from inhibitory ones, and to also identify novel microorganisms that can utilise the principal carbon sources in pyrolysis bio-oil. This study employed a phe… Show more

“…For the species-level identification, the BIOLOG TM identification system was applied (BIOLOG TM , USA) based on the utilization of carbon sources and chemical sensitivity assays in yeast identification microplate (YT) test panels (11,33). All the chemicals, reagents, and growth medium were supplied by (BIOLOG TM , USA).…”

“…Repeating Pipettor was filled by drawing up the cell suspension from the reservoir and all 96 wells were inoculated with 100 µl of the suspension. The microplate was then covered with its lid and incubated at 30 • C for 24-48 h. After incubation, the microplate was placed into the Micro Station Reader for analysis to obtain ID result and the result was given through comparison with the database using the software program MicroLog 4.20.05 (BIOLOG TM , USA) (33,34). The scope of the 96 assay reactions, coupled with sophisticated interpretation software, delivers a high level of accuracy that is comparable to molecular methods.…”

“…BioLog TM (BIOLOG™, USA) is an advanced microbial identification system that can swiftly identify species of aerobic and anaerobic bacteria, yeasts, and fungi. The latest generation redox chemistry of the BioLog TM enables testing and identification of aerobic gram-negative and gram-positive bacteria in the same test panel ( 20 ). For the species-level identification of predicted Lactobacillus sp.…”
