Identification of amino acid transport systems stimulated by FSH in rat testes

Abstract: Testes of 12-day-old rats preincubated (90 min) and incubated (60 min) with FSH in Krebs-Ringer bicarbonate buffer (KRB) increased their uptake of [14C]methylaminoisobutyric acid and [14C]aminoisobutyric acid. The hormone was ineffective in sodium-free KRB. Transport of [14C]aminoisobutyric acid in the presence of methylaminoisobutyric acid (MeAIB; 30 mmol/l) was not stimulated by FSH, and uptake of [14C]cycloleucine was not increased by FSH in any of the experimental conditions used. It was concluded that in … Show more

“…This FSH stimulatory action was independent of its effect on AC-cAMP activity and was observed using FSH from different species and sources (NHPP: ovine, porcine, and rat; Sigma-Aldrich: ovine) (Irusta and Wassermann, 1974; Pérez-Sánches and Wassermann, 1981; Cruz Curte and Wassermann, 1985; Spritzer and Wassermann, 1985; Silva and Wassermann, 1999; Silva et al, 2002). …”

“…Following the maximum hormonal effect, hyperpolarization remained in a steady state until the end of the measurement (35-45 s). For the experimental data, the dual effect of FSH on hyperpolarization, followed by depolarization, was found to be concentration-dependent as depolarization was induced by the Na+-amino acid co-transporter and involved calcium influx 12,29,31 . The model was implemented in MATLAB®, using the ode45 function to solve the differential equations running in Windows 7.…”
