Identification and expression of DoCCaMK during Sebacina sp. symbiosis of Dendrobium officinale

Abstract: Dendrobium officinale Kimura et Migo is a famous precious medicinal plant in China. Seed and seedling were cultivated with the mycorrhizal fungus Sebacina sp. CCaMK was initially cloned from D. officinale based on a SSH cDNA library of symbiotically germinated seeds with Sebacina sp. Phylogenetic analysis was performed among DoCCaMK and other CCaMKs. The particle bombardment technique was used to visualize DoCCaMK-GFP. qRT-PCR and western blot analysis were conducted to determine the tissue expression patterns… Show more

“…All the 6 selected genes related to oxidase, reductase and energy metabolism from the RNA-seq data exhibited differential expression in P. umbellatus in response to oxalic acid. The qRT-PCR protocol and the reaction procedure were all performed according to a previous study 39 . The gene expression ratio was evaluated by the comparative 2 −ΔΔCt method and the fold changes were calculated by logarithm of 2.…”

“…In recent years, various omics technologies, such as genome, transcriptome, proteome and metabolome, have been applied to elucidate the complexity of ORM interaction and provide a preliminary understanding of the establishment and signalling of mycorrhizal symbiosis in Orchidaceae. The common symbiotic signalling pathways (CSSP) involved in AM symbiosis are believed to exist in ORM ( Miura et al, 2018 , Xing et al, 2020 ). Plant roots exude hormone or flavoid compounds, and these compounds stimulate fungi to release signalling molecules; the two partners recognise each other through the receptor protein, promote successful fungal colonisation inside plant roots and further trigger the transcription regulation of mycorrhizal symbiosis-related genes ( Favre-Godal et al, 2020 ).…”

“…According to the transcriptome analysis, the DEGs of comp16933_c0, comp34626_c0 and comp32446_c0, respectively, encoding ERG6, ERG2 and ERG5 were selected for validating their expression patterns in the QR and the control groups. The PCR-amplification, the qRT-PCR protocol were all conducted using Real-time fluorescence quantitative PCR instrument (Roche, LightCycler 480), according to a previous study (Xing et al, 2013(Xing et al, , 2020.…”
