Citrus sinensis is one of the most important Vitamin C containing fruit all over the world which is badly infected by brown rot disease (BRD). The present research work was designed to isolate and characterize the pathogenic fungus responsible for BRD of C. sinensis as well as evaluation of its antagonistic control measures. The causal agent of BRD was isolated and characterized using physiological, morphological and molecular methods. The pathogenic fungus was isolated from the skin of sweet orange. The isolated fungus was grown in different nutrients media, temperature, pH, sugar concentrations and different carbohydrates. The highest growth of the fungi was observed in 5% fructose containing PD broth media at 37 0 C, pH 7.5 and 0.05gm/L NaCl concentration. PCR amplification of the isolated fungal DNA by internal transcribed spacer (ITS) region products showed approximately 650bp size of clear cut band. sequencing analysis showed that the amplified its region of the fungus (GenBank Submission# SUB5672073) has 99.80% similarities with the sequences of Pichia kudriavzevii isolate 11-577 its-1 partial sequence. In pathogenicity assay, artificially infected P. kudriavzevii fungus in lemon, malta and apple fruits showed a typical brown rot disease symptoms and the isolates showed similar size of 650bp clear band for PCR products. In antifungal activity assay, the methanol extracts of Allium sativum displayed highest 17.18±0.66 mm diameter zone of inhibition against the isolated fungus. Rhizobium leguminosarum and Alternaria spp. showed significant antagonistic effect in coculture with the isolated fungus. The present research work would be helpful to control the devastating brown rot disease of Citrus sinensis.

Rhizoctonia solani (RS) is a pathogenic fungus that affects maize (Zea mays L.) plants and causes banded leaf and sheath blight (BLSB) with severe consequences leading to significant economic losses. Contrarily, rhizobacteria produce numerous volatile organic compounds (VOCs) that help in devising the environment-friendly mechanism for promoting plant growth and stress alleviation without having physical contact with plants. In the present study, 15 rhizobacterial strains were tested for their antagonism against RS. The antagonistic potential of VOCs of the tested plant growth-promoting rhizobacteria (PGPR) strains ranged from 50% to 80% as compared to the control (without PGPR). Among these 15 strains, the maximum (80%) antagonistic activity was exhibited by Pseudomonas pseudoalcaligenes SRM-16. Thus, the potential of VOCs produced by P. pseudoalcaligenes SRM-16 to alleviate the BLSB disease in maize was evaluated. A pot experiment was conducted under greenhouse conditions to observe the effect of VOCs on disease resistance of BLSB-infected seedlings. Overall, maize seedlings exposed to VOCs showed a significant increase in disease resistance as indicated by a reduced disease score than that of unexposed infected plants. The VOCs-exposed maize exhibited lower (11.6%) disease incidence compared to the non-inoculated maize (14.1%). Moreover, plants exposed to VOCs displayed visible improvements in biomass, photosynthetic pigments, osmoregulation, and plant antioxidant and defense enzyme activities compared to the healthy but unexposed seedlings. Simultaneous application of RS and VOCs enhanced superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), phenylalanine ammonia lyase (PAL), ascorbate peroxidase (APX), and polyphenol oxidase (PPO) activities by 96.7%, 266.6%, 313.7%, 246.6%, 307%, and 149.7%, respectively, in the roots and by 81.6%, 246.4%, 269.5%, 269.6%, 329%, and 137.6%, respectively, in the shoots, relative to those of the control plants. The binding affinity of the VOCs (2-pentylfuran, 2,3-butanediol, and dimethyl disulfide) with CRZ1 and S9 protein receptors of RS was assessed by deploying in silico methods. Overall, 2-pentylfuran exhibited a binding affinity with both the selected receptors of RS, while 2,3-butanediol and dimethyl disulfide were able to bind S9 protein only. Hence, it can be deduced that S9 protein receptors are more likely the target RS receptors of bacterial VOCs to inhibit the proliferation of RS.

Diplodia gummosis disease caused by Lasiodiplodia theobromae is an economically important postharvest fruit decay that occurs on all types of citrus grown in Bangladesh. The present investigation was conducted to isolate and identify the pathogenic fungus responsible for postharvest diplodia gummosis disease (DGD) of citrus as well as the evaluation of its biological control through microbial antagonists. DGD causing pathogen was identified by physiological, morphological and molecular methods. The pathogenic fungus was isolated from the surface of postharvest lime fruits. The optimum growth of the fungi was observed in maltose, 2% NaCl, 15% sugar and pH 7 containing PDA medium at 35⁰C. PCR products of the internal transcribed spacer (ITS) region of the fungus showed approximately 650 bp size clear band in gel electrophoresis. The amplified region of the fungus showed 99.62% similarities with the sequences of L. theobromae. Artificially inoculation of the fungus in malta, musumbi, sweet orange, lime, and guava fruits showed similar size clear band and typical diplodia gummosis disease symptom. Methanol extracts of Datura metal displayed the highest inhibition (75.25%) against the isolated fungus. Non-pathogenic fungi Trichoderma viride showed the highest antagonistic efficiency followed by Neofusicoccum mangifera against the isolated fungus. The tested soil bacteria did not show significant antagonistic activity against the isolated fungus. Therefore, the DGD of citrus control system should be integrated into the overall citrus postharvest decay control system to reduce all citrus postharvest diseases and to protect fresh citrus fruit values.
