Identification and characterization of a monoclonal antibody to an antigen expressed on activated macrophages.

Abstract: A hybridoma clone secreting a monoclonal antibody, designated MA158.2, that reacts with an antigen ex pressed on lymphokine-treated macrophages was produced by fusion of mouse myeloma cells with rat spleen cells immunized against C57BL/6 peritoneal macrophages rendered tumoricidal in vitro by incubation with the lymphokine macrophageactivating factor. The specificity of the antibody for activated macrophages and lack of reactivity with histologically diverse cell types was determined by radioimmune indirect bi… Show more

“…Several monoclonal antibodies specific for murine macrophage-surface antigens have been reported (12,19,(20)(21)(22). Some of these were directed against activated macrophages (12,19,21,22).…”

“…Some of these were directed against activated macrophages (12,19,21,22). In this study we established two hybridomas: B12 monoclonal antibody reacted with all macrophages, while C14 antibody reacted well with resident and thioglycollate-, glycogen-, and peptone-elicited macrophages, but little with BCG-and glucan-elicited macrophages, which mediate LDMC with loach egg lectin and release a cytotoxin (Table 2).…”

“…Anti-NP rabbit polyclonal antibody and anti-M1 mouse monoclonal antibody were obtained by immunizing animals with hexahistidinetagged NP and M1 as previously described (23,24). Anti-c-myc (9E10) and anti-FLAG (M2) mouse monoclonal antibodies were purchased from Santa Cruz Biotechnology Inc. (Santa Cruz, CA) and Sigma-Aldrich (St. Louis, MO), respectively.…”
