We evaluated the effect of chronic tobacco smoke exposure on the function of the alveolar macrophage (AM) in mice. Tumor necrosis factor-α production of the AM triggered by lipopolysaccharides was smaller in smoke-exposed mice as compared to control mice but did not reach statistical significance (27.3 ± 4.0 vs. 34.8 ± 4.9 U/ml). The percentage of AM which did not phago-cytize latex particles in the smoke-exposed mice was significantly larger than that in control mice (33.9 ± 2.3 vs. 20.8 ± 2.1%; p < 0.05). la antigen expression of the AM was significantly larger in smoke-exposed mice (cytotoxicity index: 0.180 ± 0.033 vs. 0.038 ± 0.0118; p < O.Ol). The asialo-GMl antigen expression was similar in both groups (0.949 ± 0.007 vs. 0.961 ± 0.011). Although the precise mechanisms of these functional changes of the AM by tobacco smoke exposure are not clear, they may have some immunological effects on the alveolar space.

Murine peritoneal macrophages were preincubated with amphotericin B (AMPH) and were then stimulated with bacterial lipopolysaccharide or streptococcal preparation (OK432). These macrophages produced a large amount of tumor necrosis factor. When administered to mice, the priming activity of amphotericin B for tumor necrosis factor production in vivo was also observed.Amphotericin B (AMPH) is known to activate macrophage functions in terms of oxidative burst (17) and fungicidal activity (10). It was also reported to induce the production of a marginal level of tumor necrosis factor-a (TNF) in murine macrophages in vitro (2, 3). There is much evidence that TNF exhibits many types of immunological properties (1,4,14). This suggests that some part of the immunological or pharmacological actions of AMPH may be mediated by TNF. Efficient induction of TNF by macrophages is known to require at least two types of stimulation (7). Here, we report that AMPH induces a relatively large amount of TNF in vitro and in vivo when combined with other macrophage stimulators.C3H/HeN mice were obtained from Shizuoka Experimental Animal Corporation, Hamamatsu, Japan. Deoxycholatefree AMPH supplied from Bristol-Myers Squibb K. K., Tokyo, Japan, was used unless indicated otherwise. AMPH was dissolved in dimethyl sulfoxide (DMSO), and final concentrations of DMSO in culture media were less than 0.05%. Endotoxin contamination of all tested AMPH preparations except AMPH as Fungizone (Bristol-Myers Squibb Co.) was estimated to be less than 15 pg/i ,ug of AMPH by a Limulus assay (Endospecy; Seikagaku Industrial Co., Tokyo, Japan). Bacterial lipopolysaccharide (LPS) from Escherichia coli 0127:B8 was purchased from Difco Laboratories. A streptococcal preparation, OK432, was kindly supplied by Chugai Chemical Co., Tokyo, Japan. Antitumor mannoglucan polyalcohol (MGA) was supplied by Daiichi Pharmaceutical Co., Ltd., Tokyo, Japan. Fetal calf serum, in which contaminating endotoxin was present at less than 3 pg/ml, was purchased from Hyclone Co., Logan, Utah.Male C3H/HeN mice (6 weeks old) were injected intraperitoneally with 3 ml of 3% thioglycolate. Four days later, peritoneal cells were collected, washed twice with RPMI 1640 medium containing 5% fetal calf serum, and adjusted to 1.0 x 106 cells per ml in the same medium. Classical brown adipocytes, characterized by interscapular depots, have multilocular fat depots and are known to release excess energy. Recent studies have shown that induction of brown-like adipocytes, also referred to as beige or brite cells, in white adipose tissue (WAT) results in the release of excess energy through mitochondrial heat production via uncoupling protein 1. This has potential a therapeutic strategy for obesity and related diseases as well as classical brown adipocytes. In our previous studies, we found that artepillin C (ArtC, 10 mg/kg body weight), a characteristic constituent of Brazilian propolis, significantly induced the development of brown-like adipocytes in inguinal WAT (iWAT) of mice. Furthermore, we recently demonstrated that curcumin (Cur, 4.5 mg/kg) also significantly induced the development of brown-like adipocytes in mice. The combined administration of several food-derived factors can enhance their bioactivity and reduce their required functional doses. In this study, we showed that co-administration of Cur and ArtC at lower doses (Cur, 1.5 mg/kg; ArtC, 5 mg/kg) additively induce brown-like adipocyte development in mouse iWAT. Moreover, this induction is associated with the localized production of norepinephrine following accumulation of alternatively activated macrophages in iWAT. These findings suggest that co-administration of Cur and ArtC is significantly effective to reduce the dose and enhance the formation of brown-like adipocyte via a unique molecular mechanism.
