Identification and analysis of the β-catenin1 gene in half-smooth tongue sole (Cynoglossus semilaevis)

Abstract: β-catenin is a key signalling molecule in the canonical Wnt pathway, which plays a role in cell adhesion, embryogenesis and sex determination. However, little is known about its function in teleosts. We cloned and characterized the full-length β-catenin1 gene from half-smooth tongue sole (Cynoglossus semilaevis), which was designated CS-β-catenin1. The CS-β-catenin1 cDNA consists of 2,346 nucleotides and encodes a protein with 782 amino acids. Although CS-β-catenin1 was transcribed in the gonads of both sexes,… Show more

“…Similar results appeared in two types of medaka ( O. latipes and O. sakaizumii ), which proved Gsdf affected oestrogen production during sex differentiation 19,20 . Simultaneously, Wnt4a is a key gene in the Wnt4/β-catenin1 pathway that regulates gonad development/differentiation 53,54 . CS - wnt4a was also upregulated after RNAi, although the mechanisms remain elusive.…”

“…In C. semilaevis , ovarian differentiation can be observed earlier than the testicular differentiation. Histological observations show that gonadal differentiation begins at ~ 62 days post hatching (dph) [ 24 ]. At this stage, primordial germ cells (PGCs) begin mitosis rapidly to form clusters of oocytes.…”

“…Both wnt4a KO ( Kossack et al, 2019 ) and Wnt antagonist dkk1 overexpression cause testis development ( Sreenivasan et al, 2014 ), confirming a key role of the Wnt / β - catenin pathway in ovarian differentiation in zebrafish. Different chemical inhibitors of the Wnt / β - catenin pathway have been used in other fish species, resulting in downregulation of pro-ovarian genes such as cyp19a1a or foxl2 in tongue sole ( Zhu et al, 2017 ) and carp ( Wu et al, 2019 ) or upregulation of pro-testis gene dmrt1 in tilapia ( Wu et al, 2016 ). This suggests a conservation of the Wnt / β - catenin pathway functions in ovarian differentiation in teleosts, although each actor of the pathway may not play the same part, depending on the species.…”
