Background: Endometrial carcinoma (EC) is the primary cause of death associated with cancer globally. Thus, the possible molecular mechanism of EC needs further exploration. Up-frameshift protein 1 (UPF1) is an ATPase depending on RNA/DNA and RNA helicase depending on ATP. Long noncoding RNA (lncRNA) plasmacytoma variant translocation 1 (PVT1) was dysregulated in diverse diseases. Methods: qRT-PCR and Western blot were applied to detect UPF1 and PVT1 in EC. CCK-8, colony formation, and Transwell assays were used to test the effects of UPF1/PVT1 on cell proliferation and migration. Cells were cultured with actinomycin D to observe mRNA stability, and RNA immunoprecipitation assay was applied to verified the relationship between UPF1 and PVT1. Glucose consumption and lactate generation were measured when cells were transfected with siRNA. Results: Results demonstrated that the expression of UPF1 exhibited a remarkable decrement in EC tissues relative to that in non-tumor tissues. Subsequent functional experiments suggested that UPF1 decrement stimulated EC cells to grow and migrate. Moreover, UPF1 was discovered to be linked to PVT1 and had an inverse correlation with PVT1. Besides, PVT1 expression affected EC growth and migration, and PVT1 decrement alleviated the influence of UPF1 decrement on EC growth and migration and strengthened glycolysis in EC. Conclusion: In this study, we found that UPF1 was down-regulated in EC tissues, and UPF1 might exert its role by regulating the expression of PVT1.

Background: The most common histological subtypes of esophageal cancer are squamous cell carcinoma (ESCC) and adenocarcinoma (EAC). It has been demonstrated that non-marginal differences in gene expression and somatic alternation exist between these two subtypes; consequently, biomarkers that have prognostic values for them are expected to be distinct. In contrast, laryngeal squamous cell cancer (LSCC) has a better prognosis than hypopharyngeal squamous cell carcinoma (HSCC). Likewise, subtype-specific prognostic signatures may exist for LSCC and HSCC. Long non-coding RNAs (lncRNAs) hold promise for identifying prognostic signatures for a variety of cancers including esophageal cancer and head and neck squamous cell carcinoma (HNSCC). Methods: In this study, we applied a novel feature selection method capable of identifying specific prognostic signatures uniquely for each subtypethe Cox-filter methodto The Cancer Genome Atlas esophageal cancer and HSNCC RNA-Seq data, with the objectives of constructing subtype-specific prognostic lncRNA expression signatures for esophageal cancer and HNSCC. Results: By incorporating biological relevancy information, the lncRNA lists identified by the Cox-filter method were further refined. The resulting signatures include genes that are highly related to cancer, such as H19 and NEAT1, which possess perfect prognostic values for esophageal cancer and HNSCC, respectively. Conclusions: The Cox-filter method is indeed a handy tool to identify subtype-specific prognostic lncRNA signatures. We anticipate the method will gain wider applications.

Abstract Background In recent years, long noncoding RNAs (lncRNAs) have been reported frequently to play important roles in specific cancers, including high-grade serous ovarian cancer (HGSOC). PVT1 is an important oncogenic lncRNA highly expressed in various cancers. However, little is known about the role of PVT1 in HGSOC and underlying mechanisms. Methods The expression levels of PVT1 and HER2/3 in HGSOC tissue and adjacent normal tissue were determined by qRT-PCR. MTT and transwell assays were used to identify the effects of PVT1 cell proliferation and migration respectively. Dual-luciferase reporter assay and RIP experiment were carried out to verify target genes of PVT1. ChIP experiment was used to identify that HER2 was transcription factor of PVT1. Results Our results showed that PVT1 expression was up-regulation in human HGSOC specimens and promoted ovarian cancer cell proliferation and migration. We further validated that HER2 was a direct transcription factor for facilitating PVT1 expression. In return, PVT1 enhanced HER2 transcript stability. Moreover, bioinformatics analysis and dual luciferase reporter assay results uncovered that PVT1 functioned as a competing endogenous RNA (ceRNA) for miR-1301-3p to promote cell proliferation and migration by increasing HER3 expression. Conclusions Taken together, our results showed that PVT1, controlled by HER2, elevated HER2 and HER3 expression to promote HGSOC progression. Thus, PVT1 can be regarded as a vital diagnostic biomarker for HGSOC and a potential novel therapeutic target.
