Ishige sinicola extract stimulates osteoblast proliferation and differentiation via the bone morphogenetic protein 2/runt-related gene 2 signalling pathway

Abstract: Osteoporosis is one of the most common bone diseases, occurring due to an imbalance between bone formation and bone resorption. The aim of this study was to investigate the effects of Ishige sinicola, a brown alga, on osteoblast differentiation through the activation of the bone morphogenetic protein 2 (BMP-2)/runt-related transcription factor 2 (Runx2) signalling pathway in MC3T3-E1 cells. A cell proliferation assay, alkaline phosphatase (ALP) activity assay, alizarin red staining, and expression analysis of … Show more

“…Similarly, the extract of Hizikia fusiforme, a brown alga, has been reported to stimulate skeletal activity in zebrafish, ovariectomized mice, and calvarial mouse bones [38]. We previously suggested that ISE promotes bone formation through osteoblastic cells by stimulating osteoblast marker gene expression [22]. Consistent with these results, the present study demonstrated that ISE could be a beneficial therapeutic agent which prevents OVX-induced bone loss.…”

“…I. sinicola was reported to have antiinflammatory, antibacterial, and hair growth-promoting activities [20,21]. In addition, our previous study demonstrated that the I. sinicola extract (ISE) affects osteoblast differentiation via the bone morphogenetic protein 2/runt-related gene 2 signaling pathway [22]. However, its protective effects against bone loss induced by estrogen deficiency and osteoclastogenesis have not yet been reported.…”

“…In a study focusing on determining the mechanism of hormone estrogenic effects in the cell culture, it was reported that the use of the BMP antagonist and its receptor inhibition reduced harmine effects in increasing ALP activity and the growth and differentiation of bone cells (39). Harmine also increases the expression of Runx2 and Osterix mRNA, which are the main transcription factors in differentiating osteoblasts (40).…”

“…Gene expression profile data revealed an increase in the expression of the canonical Wnt-associated genes, Ctnnb1, Wls, Wnt2b, and Wnt4 after TCQA treatment [40]. Ishige sinicola (IS) is a brown alga that exhibits various activities, including osteoblastic bone formation [150] and anti-inflammatory effects [151]. IS treatment for 3 weeks increased hair-fiber length in rat vibrissa follicles and induced anagen progression of the hair shaft [41].…”
