Pistacia integerrima leaf galls are used in several traditional medicines to cure many diseases such as diarrhea, asthma, fever, cough, vomiting, and hepatitis. The main goal of the present investigation was to assess the antidiarrheal effect of the Pistacia integerrima extracts/fractions and four isolated flavonoid compounds (1−4) on mice. An in vivo assay involving castor-oil-induced diarrhea was used to evaluate the antidiarrheal potential of extracts/fractions at 100, 200, and 400 mg/kg p.o., as well as isolated compounds at 5, 10, and 20 mg/kg p.o. Pretreatment of mice with extracts/fractions significantly attenuated castor-oil-induced diarrhea in a dose-dependent manner. Among all crude extracts and fractions, the ethyl acetate extract was the most effective with 100% protection against diarrhea followed by chloroform (75% protection) at 400 mg/kg p.o. Although all the isolated compounds exhibited strong antidiarrheal activity, isolated compounds 1 and 4 demonstrated 100% protection against diarrhea. Moreover, docking models were performed using the Molecular Operating Environment (MOE) and AutoDock software and suggested that the extracts and isolated compounds exert antidiarrheal activity by inhibiting mu-opioid and delta-opioid receptors. Therefore, our finding affords a strong pharmacological basis for the traditional use of P. integerrima galls in the treatment of diarrhea.

Micromeria biflora, a traditional medicinal plant, is extensively used for treating various painful conditions, such as nose bleeds, wounds, and sinusitis. A phytochemical investigation of the chloroform fraction of Micromeria biflora led to the isolation of salicylalazine. Salicylalazine was assessed in vivo for analgesia, muscle relaxation, sedative, and anti-inflammatory properties, as well as in vitro for COX-1/2 inhibition activities. It was assessed against a hot plate-induced model at different doses. The muscle relaxant potential of salicylalazine was evaluated in traction and inclined screening models, while sedative properties were determined using an open-field model. The anti-inflammatory potential of salicylalazine was assessed in histamine and carrageenan-induced paw edema screening models. Salicylalazine exhibited significant analgesic potential in a dose-dependent manner. In both screening models, an excellent time-dependent muscle-relaxation effect was observed. Salicylalazine demonstrated excellent sedation at high doses. Its anti-inflammatory activity was determined through the initial and late phases of edema. It exhibited anticancer potential against NCI-H226, HepG2, A498, and MDR2780AD cell lines. In vitro, salicylalazine showed preferential COX-2 inhibition (over COX-1) with an SI value of 4.85. It was less effective in the initial phase, while, in the later phase, it demonstrated significant effects at 15 and 20 mg/kg doses compared with the negative control. Salicylalazine did not exhibit cytotoxicity in the MTT assay, preliminarily indicating its safety.

The synthesis of metal nanoparticles by using plant extracts is previously explored in phytomedicines. Nanobiotechnology has many applications, including cosmetic, packing, coating, biomedicine, and enhanced biological activity. Keeping in view the importance of Pistacia chinensis, its gold nanoparticles (AuNPs) have been synthesized by the eco-friendless and cost-effective method. In this study, the synthesized nanoparticles were characterized by advanced techniques such as UV-visible spectroscopy, Fourier transform infrared (FT-IR), and atomic force microscope (AFM) analysis. The biological activities of these synthesized nanoparticles were examined in vitro by measuring the enzymatic inhibition potential on urease and carbonic anhydrase and in vivo by determining the analgesic and sedative activities. The UV spectrum indicated various peaks at the range of 530-550 nm, showing nanoparticles formation. The FT-IR spectroscopy of the extracts and AuNPs indicated the presence of NH, C═N, and N═O in the extract involved in the nanoparticles synthesis. The size of nanoparticles was determined by AFM analysis. The AFM showed that the nanoparticles range from 10 to 100 nm and are almost spherical in shape. The synthesized AuNPs exhibited significant urease inhibition potential with an IC50 value of 44.98. Similarly, the nanoparticles exhibited good carbonic anhydrase inhibition with an IC50 value of 53.54 against acetazolamide having IC50 0.13. Pistacia chinensis extract and its AuNPs exhibited excellent attenuation p < 0.01 in acetic acid-induced writhing model at a dose of 15 mg/kg. The synthesized nanoparticles showed a significant sedative effect p < 0.001 compared to the standard drug. This research work has developed a green method to synthesize nanoparticles by using Pistacia chinensis extract and directed the researcher to purify active phytochemicals from Pistacia chinensis involved in nanoparticles synthesized.
