In vitroOrganogenesis and Plantlet Formation ofPaulownia elongata

Abstract: SYNOPSISShoot tips, nodes, in~ernodes and leave, sderived from aseptically produced seedlings of Paulownia elongata were cultured on Murashige and Shoog medium (MS) supplemented with NAA and BA. Medium containing a 1 mg/l NAA and 0,5 mg~f BAgave,th.e best shoot production from leafcallus (50 shoots per 25 mm 2 segment). Sh~ot tips cultured on medium containing l.mg/f BA gave rise to a 9,7-fold increase in shoot production in 30 days. The shoots prod';lced could be rooted e.lther on MS medium without growth regu… Show more

“…Ratio of NAA/BAP was 1/5 in those works. The results in this study corroborated those of Rao et al (1996) and Chang and Donald (1992) indication that is the increased ratio of BAP and decreased ratio of NAA was significant to induce shoot for all explants (2, 6). Song et al (1990) noticed that ratio of shoot multiplication from P. catalpifolia meristems was enhanced by increasing concentrations of BAP up to 8.0 mg l -1 (7).…”

“…Rao et al (1996) reported that optimum shoot production from petioles of P. fortunei was obtained when MS medium supplemented with 4.5 mg.l -1 BAP and 0.8 mg.l -1 NAA was used (2). Chang and Donald (1992) used MS medium combined with 0.1 mg l -1 NAA + 0.5 mg l -1 BAP for regeneration of shoots from leaf callus of P. Elongata (6). Ratio of NAA/BAP was 1/5 in those works.…”

“…In vitro propagation techniques have been used for regenerating plants from various explants of Paulownia (Marcotrigiano and Stimart, 1983 [8] ; Jagannathan and Marcotrigiano, 1986 [9] ; Ni and Ma, 1982 [10] ; Chang and Donald, 1992 [11] ). To date, there has been no report of shoot production from the nodes of young seedlings of P. elongata, an economically important species in this genus.…”

“…Furthermore, a vegetative propagation system based on adventitious shoot production could be integrated with genetic engineering. In vitro propagation techniques have been used for several species as well as two hybrids of Paulownia (Marcotrigiano and Stimart 1983;Jagannathan and Marcotrigiano 1986;Ni and Ma 1982;Chang and Donald 1992). Although plant regeneration with direct and indirect organogenesis from various explant sources of Paulownia species have already been reported, to date there is only one report on indirect somatic embryo formation from P. tomentosa (Radojevic 1979).…”
