In‐vitro assays of polymer‐coated stents eluting platelet glycoprotein IIb/IIIa receptor monoclonal antibody

Abstract: The monoclonal antibody (mAb) to the platelet glycoprotein (GP) IIb/IIIa receptor has potent antiplatelet and antithrombotic characteristics shown to reduce thrombus-related major complications after coronary angioplasty. This mAb can be incorporated in drug-eluting stents capable of releasing single or multiple bioactive agents into the bloodstream and surrounding tissues. Stents eluting the monoclonal mouse anti-human platelet glycoprotein IIb/IIIa antibody SZ-262 were tested for their effectiveness in impro… Show more

“…SZ‐21 (Jiangshu Institute of Hematology, Suzhou, China), the mAb against platelet GP IIIa, can inhibit platelet aggregation after the antibody is absorbed by the PLLA polymer coating on the surface of the stents 13. As shown in the present experimental results, the SZ‐21‐eluting stent can inhibit platelet adherence and aggregation, thus preventing stent thrombosis in vivo .…”

“…The mAb‐eluting stents were prepared by passive absorption method. The presence of the PLLA polymer coating and the mAb on the entire surface of the stent was examined using fluorescence staining and immunofluorescence technique, respectively 13. The adsorption characteristics of the stents exposed to the mAb were tested by radioisotope technique with 125 I‐labelled SZ‐21.…”

“…The SZ‐21 has the ability to block the GP IIb/IIIa receptors on the platelets, preventing the formation of fibrinogen bridges, the final common pathway in platelet thrombus formation 30. Although the chitosan is often used as a procoagulant material, the hemostatic role of chitosan is indirect, through its nonspecific adsorption of blood cells, rather than a direct activation of the blood coagulation system 46.…”
