ObjectiveThe objective of this study was to evaluate the antimicrobial activity of Mineral trioxide aggregate e Portland cement against some selected cariogenic bacteria. Methods Wells were made of approximately 0.5 mm in diameter, in solid culture media and immediately filled with cement. Twelve samples of each material were obtained for the realization of the agar diffusion method. These samples were tested with Streptococcus mutans (ATCC 700610), Enterococcus faecalis (ATCC 29212), Lactobacillus acidophilus (UFV) and Lactobacillus casei (UNICAMP). Petri plates containing Tripticase soy Agar (TSA) were used for the growth of Streptococcus mutans and Enterococcus faecalis, and plates of Man Rogosa & Sharpe Agar (MRS) for the Lactobacillus acidophilus and Lactobacillus casei. After 48 hours of incubation, it was made the measurement of inhibition halos with the aid of manual caliper. ResultsThe diameters obtained to MTA (2.50 ± 0.00;2,92 ± 0.63; 2.58 ± 0.38), PC (1.17 ± 0.29; 2.00 ± 0.43; 1.33 ± 0.14) and calcium hydroxide cement (3.83 ± 0.29; 3.00 ± 0.00 ± 2.58 ± 0.72) ResultadosOs diâmetros obtidos do MTA (2.50 ± 0.00; 2,92 ± 0.63; 2.58 ± 0.38), CP (1.17 ± 0.29; 2.00 ± 0.43; 1.33 ± 0.14) e cimento de hidróxido de cálcio (3.83 ± 0.29; 3.00 ± 0.00 ± 2.58 ± 0.72) foram expressos em milímetros e submetidos ao teste de Tukey (p<0.05). Conclusão METHODSThe tested materials were: MTA (Angelus Soluções Odontológicas, Londrina, Brasil), Portland cement (CP II -F32, ITAPESSOCA AGRO-INDUSTRIAL S.A., Goiana, Brasil) and calcium hydroxide paste (Reagen TM).The antimicrobial activity of the materials was evaluated by the Agar diffusion method against four different strains.The strains used for analysis were Streptococcus mutans (ATCC 700610), Enterococcus faecalis (ATCC 29212), Lactobacillus acidophilus and Lactobacillus casei isolated by the Laboratory of Microbiologic Science (Federal University of Viçosa -MG (UFV) and University State of Campinas -UNICAMP, SP, Brazil), respectively.Of each bacterial culture tested, it was held a concentration suspension of approximately 5x108 of colonies per mL, corresponding to 0.5 of Mc Farland em range of sterile medium Trypticase Soy Browth (TSBMerck). The Streptococcus mutans and Enterococcus faecalis suspensions were inoculated with spread plate in the middle of Sodium Tioglicolate (Merck) culture and Lactobacillus and Lactobacillus acidophilus casei were inoculated in the culture medium Man Rogosa & Sharpe Agar (MRS-HIMEDIA). Subsequently, the plates were incubated at 35 ± 1°C for 24/48h and the halos of inhibition were expressed in millimeters, using a caliper manual.A total of 12 plates were employed, each microorganism was tested in triplicate. This study evaluated in vitro the antibacterial activity of 4 root canal filling materials for primary teeth -zinc oxide and eugenol cement (ZOE), Calen paste thickened with zinc oxide (Calen/ZO), Sealapex sealer and EndoREZ sealer -against 5 bacterial strains commonly found in endodontic infections (Kocuria rhizophila, Enterococcus faecalis, Streptococcus mutans, Escherichia coli and Staphylococcus aureus) using the agar diffusion test (agar-well technique). Calen paste, 1% chlorhexidine digluconate (CHX) and distilled water served as controls. Seven wells per dish were made at equidistant points and immediately filled with the test and control materials. After incubation of the plates at 37 o C for 24 h, the diameter of the zones of bacterial growth inhibition produced around the wells was measured (in mm) with a digital caliper under reflected light. Data were analyzed statistically by analysis of variance and Tukey's post-hoc test (α=0.05). There were statistically significant differences (p<0.0001) among the zones of bacterial growth inhibition produced by the different materials against all target microorganisms. K. rhizophila was inhibited more effectively (p<0.05) by ZOE, while Calen/ZO had its highest antibacterial activity against E. faecalis (p<0.05). S. mutans was inhibited by Calen/ZO, Sealapex and ZOE in the same intensity (p>0.05). E. coli was inhibited more effectively (p<0.05) by ZOE, followed by Calen/ZO and Sealapex. Calen/ZO and ZOE were equally effective (p>0.05) against S. aureus, while Sealapex had the lowest antibacterial efficacy (p<0.05) against this microorganism. EndoREZ presented antibacterial activity only against K. rhizophila and S. aureus. The Calen paste and Calen/ZO produced larger zones of inhibition than 1% CHX when the marker microorganism was E faecalis. In conclusion, the in vitro antibacterial activity of the 4 root canal filling materials for primary teeth against bacterial strains commonly found in endodontic infections can be presented in a decreasing order of efficacy as follows: ZOE>Calen/ZO>Sealapex>EndoREZ.

A presença de bactérias nos túbulos dentinários, foraminas apicais e no delta apical é considerado como falha de procedimento. Alguns estudos revelam que após o tratamento endodôntico em sessão única, bactérias ainda permanecem em estruturas dentais. O objetivo do presente estudo foi avaliar o tratamento endodôntico em uma ou duas sessões, usando hidróxido de cálcio associado ao paramonoclorofenol canforado (PMCC) como "curativo de demora". Foram utilizados quatro cães, onde dois animais foram submetidos a tratamento em sessão única e os outros em duas sessões. Foram utilizados no estudo 40 dentes: segundo e terceiro pré-molares superiores e segundo, terceiro e quarto pré-molares inferiores divididos em dois grupos.
