In vitro and in vivo studies with tetra‐hydro‐jasmonic acid (LR2412) reveal its potential to correct signs of skin ageing

Abstract: The in vitro and in vivo data demonstrate that based on its multiple interactions within human skin, LR2412 has potential to partially correct the signs of ageing in intrinsically and photoaged skin.

“…Subjects' perception confirmed the good safety profile of Retinol/LR2412: subjects considered that the daily use of Retinol/LR2412 was safer than that of tretinoin 0.025%. These results are not unexpected as a reason for the good safety profile Retinol/LR2412 may reside in the anti‐inflammatory characteristics of LR2412 according to the LR2412 ability of inhibiting the synthesis of mediators such as IL8 and PGE2 involved in inflammation and irritation.…”

“…LR2412 has shown in vitro and in vivo its efficacy in counteracting signs of skin aging. 13,14 Indeed, LR2412 enhances in vitro the deposit of hyaluronic acid (HA) in the basal and suprabasal layers of the epidermis. LR2412 stimulates the hyalurosome and increases HA Synthases 2 and 3 and CD44 (HA receptor) expression.…”

“…3D models offer an attractive alternative to current 2D in vitro and in vivo assays as they provide a more in vivo -like context than 2D assays and eliminate species difference by enabling drug testing directly in a human model system. For example, in a 3D LSE, treatment with a jasmonic acid derivative, LR2412, rescued the loss of ECM proteins observed with ageing, including collagen IV, indicating that the compound possesses ‘anti-ageing’ properties ( Tran et al, 2014 ). Plant extract 1201, which has senolytic properties, blocked the detrimental effects of the SASP from stress-induced premature senescence (SIPS) HDFs in a human LSE causing rescue of the senescent cell dependent reduction in epidermal thickness and impaired differentiation of keratinocytes ( Lammermann et al, 2018 ).…”

“…in vitro model in a keratinocyte migration situation called "migration model" (partially mimicking the suction blister in vivo clinical model) and compared it to the existing 3D RealSkin ® regeneration standard model [6] referred here as "standard model." The standard model allows vertical regeneration (homoeostasis maintenance) to being studied while migration model introduces the additional step of re-epithelialization as observed in the phase II of wound-healing process.…”
