In vitro and in vivo evaluation of PEGylated nanoparticles of bendamustine for treatment of lung cancer

Abstract: The purpose of this study was to develop PEGylated nanoparticles of bendamustine (BM) to improve therapeutic efficiency of drug and reduce the side-effects. The nanoparticles were prepared by a modified diffusion-emulsification method. The particle size and zeta potential of optimized BM-loaded PEGylated NPs were found to be 256 nm and -29.1 mV. The in vitro release showed biphasic behavior, with initial burst release followed by slow sustained delivery. The anti-tumor activity was determined using the A- 549 … Show more

“…The results showed that the cytotoxicity of epirubicin combined with dihydroartemisinin at various concentrations was stronger than that of epirubicin alone, whereas the survival rate of A549 cells was less affected by dihydroartemisinin alone (Figure 5 To evaluate the inhibition of tumor migration by liposomal formulations, a transwell migration experiment was performed. The results showed that A549 cell migration was significantly inhibited in the presence of liposomes modified with dihydroartemisinin (Figure 6(A modified epirubicin-dihydroartemisinin liposomes, wound healing was inhibited, further validating the inhibition of tumor migration (Figure (7)). Inhibition of tumor cell adhesion to the basement membrane is an important approach to inhibit tumor metastasis [38].…”

“…Presently, gene therapy and immunotherapy have the potential to improve the outcome of NSCLC [6]. However, due to the lack of clinically relevant techniques to assess the expression of target genes in patients, the cost of immunotherapy remains high compared to conventional cytotoxic therapies [7]. Therefore, chemotherapy is still an important antitumor strategy for those with inoperable NSCLC [8].…”

“…1). 28,29 Similarly to benzimidazole hybrids, 2-aminothiazoles are also a familiar group of heterocyclic compounds that exhibit a broad range of biological activities, and their advantages as medicines have been well established. 30,31 For example, the thiazole nucleus is an integral component (main core) of all available penicillins, which have transmuted the therapy of bacterial diseases.…”

“…It has become the most prevalent cause of cancer-related deaths in men and the second most common in women. Lots of data show that the inheritance of cancer genes, long-term heavy smoking and serious air pollution are considered to be related to the occurrence of lung cancer [1,2]. Non-small-cell lung cancer (NSCLC), a kind of frequently occurring lung cancer, is characterized by short proliferative cycle, strong metastatic activity and high fatality rate.…”
