In Vitro and In Vivo Characterization of the New Analgesic Combination Beta‐Caryophyllene and Docosahexaenoic Acid

Abstract: Beta-caryophyllene (BCP) and docosahexaenoic acid (DHA) are components of several plants with documented anti-inflammatory and analgesic effects in animal pain models. In the present study, in vitro and in vivo tests were carried out to evaluate their effects, alone or in combination, during long-lasting administration in a model of persistent pain. IR spectra of the two compounds were obtained to determine their chemical stability and then in vitro toxicity was evaluated in fibroblasts and astrocytes. In the … Show more

“…For this purpose, Fiorenzani et al. studied the antinociceptive activity of BCP in mixture with docosahexaenoic acid (DHA). DHA is a member of omega‐3 polyunsaturated fatty acids (PUFAs) and well‐known anti‐inflammatory mediator .…”

“…One can hypothesize that better analgesic effects may be obtained when BCP is used in combination with other natural agent(s) of desired properties. For this purpose, Fiorenzani et al [61] studied the antinociceptive activity of BCP in mixture with docosahexaenoic acid (DHA). DHA is a member of omega-3 polyunsaturated fatty acids (PUFAs) and well-known anti-inflammatory mediator [62].…”

“…In this work we used an essential oil – BCP – and a polyunsaturated fatty acid – DHA – to investigate anti‐inflammatory effects. Only one study in the literature combined both substances applied in vitro and in vivo to test the analgesic activity of this combination, revealing potent antinociceptive effects from BCP‐DHA, but they did not present an explanatory hypothesis for this synergistic activity …”

“…However, there is little research on the effects of the association of these two substances. It is only known that BCP and DHA together can present potent analgesic action . The objective of this work was therefore to determine the anti‐inflammatory profile of β BCP associated with DHA in a mouse sepsis model induced by intraperitoneal inoculation of Staphylococcus aureus .…”
