Background/Aims: Cytokines IL-2 and IL-8 both participate in immune regulation. However, the relationship between polymorphisms in these two cytokines and the risk of acquired aplastic anemia (acquired AA) has not been explored. Methods: We selected five SNPs including rs11575812, rs2069772 and rs2069762 of IL-2, rs2227306 and rs2227543 of IL-8. SNaPshot genotyping was used to test the genotypes of IL-2 and IL-8 polymorphisms in a population of 101 acquired AA patients and 165 healthy controls. Results: The rs2069762 G allele appeared to be a protective mutation, but no significant differences were found in other four SNPs. We also found that rs2069762 had an impact on the transcriptional regulation. Conclusions: It could be assumed that the rs2069762 polymorphism might reduce the risk of acquired aplastic anemia, while the remaining four SNPs might not contribute to susceptibility to acquired AA in a Chinese population.

Objective: T helper 17 (Th17) is believed to play a crucial role in pathophysiology of systemic lupus erythematosus (SLE). Our study aimed to detect the potential association of IL-21 and IL-23R polymorphisms with susceptibility to SLE. Method: Two hundreds and ten patients with SLE and the same number of matched controls were genotyped by polymerase chain reaction to detect two polymorphisms of IL-21 and one polymorphism of IL-23 receptor (IL-23R). Clinical data was collected from patients’ archives. Results: IL-21 polymorphisms (rs4833837 C/T and rs2055979 G/T) did not show any association with the disease susceptibility (p= 0.54 and p= 0.49, respectively). Comparison of patients with rs2055979GG and T allele carriers showed higher frequency of rs2055979GG genotype in patients with pleurisy compared to the ones without pleurisy (p=0.004). In addition, categorization of IL-23R rs10889677 to GG and T allele carriers (TT+TG) showed an association between TT+TG genotypes and susceptibility to SLE (p=0.0002). Moreover, a significant association was found between the inheritance of rs10889677GG+TG genotypes and presence of mucosal ulcers in SLE patients (p=0.01).Discussion: The results showed that the lower IL-21 producer rs2055979GG genotype was associated to pleurisy. Lower IL-21 serum levels in patients with rs2055979GG genotype may lead to more frequent pulmonary infections and an increased risk of pleurisy. Association of high+ intermediate IL-23R producer genotypes (rs10889677TT+TG) with SLE may be explained by possible overactivation of Th17 cells. Higher activity of Th17 cells may also explain the over representation of mucosal ulcer in patients with high producer genotype of IL-23R.

Aim of the studyThe aim of this study was to investigate the role TNF-α, IL-2, and IL-2RB variants in psoriasis (Ps) and to evaluate the association between these variants and clinical features.Material and methodsA total of 74 psoriatic patients and 74 healthy individuals were genotyped for these variants by PCR and/or RFLP.ResultsThe AA genotype of TNF-α (–308) was significantly more common in the patients (p = 0.013). TNF-α (–238) AA genotype was significantly increased in the patients (p = 0.028), while the GG genotype was decreased in the patient group, compared to the controls (p = 0.016). IL-2 (–330) variant GG and TT genotype was more common in the patients (p = 0.037, p = 0.009, respectively), while IL-2 (–330) GT genotype was increased in the control subjects (p = 0.001). IL-2 (–330) GG genotype frequency was significantly decreased (p = 0.021) and the TT genotype frequency was significantly increased among patients with psoriatic arthritis in comparison with Ps patients (p = 0.014). IL-2RB TC genotype frequency was significantly decreased and TT genotype frequency was significantly increased in the patients with positive family history of Ps compared to those who had a negative family history (p = 0.017, p = 0.014, respectively). Also, IL-2RB CC genotype was significantly increased among the patients with late-onset Ps in comparison with the early onset Ps group (p = 0.009). The frequency of IL-2 (–330) TT genotype was significantly higher in mild Ps patients than moderate-severe patients (p = 0.043).ConclusionsOur data suggest a potential role of these genes as candidate genes for susceptibility to Ps in a Turkish cohort.
