Hypoxia Increases Thyroid Cancer Stem Cell‐Enriched Side Population

Abstract: IntroductionHypoxic stress is a feature of rapidly growing thyroid tumours. Cancer progression is thought to be driven by a small population of tumour cells possessing stem cell properties. Hypoxia-inducible factors (HIFs) are important mediators of hypoxia. Both HIF-1alpha and HIF-2alpha have been reported to be expressed in thyroid cancers. There is growing evidence that the HIF pathway plays a significant role in the maintenance of thyroid cancer stem cells (CSC).MethodologyWe have isolated thyroid CSC from… Show more

“…Both HIF-1α and HIF-2α were reported to be expressed in TC (49). Moreover, a growing evidence points out that hypoxia plays a significant role in the maintenance of thyroid cancer stem cells (CSC) (50). General hypoxia due to diminished vascularization is a well-known characteristic of aging tissues, possibly due to endothelial dysfunction as well decreased function of endothelial progenitor cell function (51,52).…”

“…Hypoxia also participates in the progression of cancer at further stages of the process. In particular, hypoxia is important in the functioning of cancer stem cells (CSCs) [ 50 , 51 , 52 , 53 , 54 , 55 ], which increase the resistance of the tumor to anticancer therapy. Hypoxia also causes cancer cell migration, invasion and metastasis, partly due to hypoxia causing the epithelial-to-mesenchymal transition (EMT) [ 55 , 56 , 57 , 58 , 59 ].…”

“…111 Many studies have reported that HIFs play a critical function in the regulation of the CSC phenotype. [112][113][114][115][116][117] The cells are cultured in the 2D culture system uniform contact with oxygen, which cannot create a continuous oxygen concentration gradient. Thus, the 2D cell culture system does not mimic a hypoxic environment similar to in vivo conditions.…”
