Hypoxia and Reoxygenation-Induced Injury of Renal Epithelial Cells: Effect of Free Radical Scavengers

Abstract: ABSTRACT-The aim of this study was to characterize injuries of LLC-PK1 and MDCK cells exposed to hypoxia and reoxygenation. Exposure of LLC-PK1 cells to hypoxia reduced the ATP contents and increased the leakage of lactate dehydrogenase (LDH), but MDCK cells had no such injuries. Hypoxia-reoxygenation of LLC-PK1 cells dramatically increased LDH leakage, which was suppressed by free radical scavengers, N N diphenyl p-phenylenediamine, superoxide dismutase and N. N dimethylthiourea. These results suggest that us… Show more

“…The extent to which SOD or catalase are internalized has been questioned (Yonehana and Gemba, 1995;Doroshow 1986). In rat hepatocytes incubated with SOD, intracellular SOD activity was increased about 4-fold (Kyle et al, 1988).…”

“…The decrease in cell viability with DMTU was not due to toxicity of the scavenger itself since the toxicity was not seen in cells incubated with DMTU alone. DMTU has been used with opossum kidney cells (Park et al, 2003) and LLC-PK 1 cells (Yonehana and Gemba, 1995) with no evidence of toxicity. The interaction between PAP and DMTU is perplexing and will require further study.…”

“…Among these, it has been widely accepted that reactive oxygen species participate in the pathogenesis of ischemic acute renal failure (2,3). Therefore, investi gations of amelioration or prevention of cellular injury after renal ischemia through manipulation of free radi cal production have received much attention and indi cated that antioxidants have therapeutic potential (4,5).…”

“…Although no signi®cant suppression was observed within the tested concentration, the level of LDH leakage tended to decrease as the concentrations of caffeic acid, rosmarinic acid and lithospermic acid increased, suggesting that these compounds may have activity at higher concentrations. Yonehana & Gemba (1995) found that the intracellular level of the antioxidant glutathione was signi®cantly decreased under hypoxic conditions in LLC-PK 1 cells. Snowdowne et al (1985) and Kribben et al (1994) reported that hypoxia caused an increase of intracellular calcium in a kidney-derived cultured cell line, LLC-MK 2 , and also in isolated renal tubules.…”
