Hydrogen-Peroxide Synthesis and LDL-Uptake Controls Immunosuppressive Properties in Monocyte-Derived Dendritic Cells

Abstract: Background and Aims: Induction of myeloid-derived suppressor cells (MDSC) is a critical step in immune cell evasion by different cancer types, including liver cancer. In the liver, hepatic stromal cells orchestrate induction of MDSCs, employing a mechanism dependent on hydrogen peroxide (H2O2) depletion. However, the effects on monocyte-derived dendritic cells (moDCs) are unknown. Methods: Monocytes from healthy donors were differentiated to moDCs in the presence of extracellular enzymatic H2O2-depletion (here… Show more

“…These changes are significant, implying that all the treatments elicit a negative effect on neutrophil level. Monocytes play crucial roles in innate and adaptive immunity during inflammation [48][49][50]. The results of the present study revealed a low count of monocyte cells, which could result from immunosuppression and bone marrow failure and/or damage [51][52].…”

“…Apart from LDs, lipoprotein metabolism also impacts DC function. Immature moDCs were shown to display notably increased NADPH-oxidase-driven H 2 O 2 -production and LDL uptake ( 81 ). These features contributed to the immunosuppressive function of immature moDCs, whereas blocking LDL uptake restored their maturation capacity and attenuated their immunosuppressive properties.…”

“…Six of the papers focus on the role of specific cell types in tumor immune evasion. Among these, the research paper by Menzner et al [ 9 ] reported a novel axis between hydrogen peroxide and low-density lipoproteins-mediated axis in immunosuppressive abilities of monocyte-derived dendritic cells. The review by Demkow [ 10 ] underlined the multifaceted crosstalk between cancer and neutrophil extracellular traps during tumor progression.…”
