Human Serum With High Neutralizing Antibody Titers Against Both Zika and Dengue Virus Shows Delayed In Vitro Antibody-Dependent Enhancement of Dengue Virus Infection

Abstract: Zika virus infection in a dengue virus–naïve subject was associated with the induction of high levels of cross-reactive binding antibodies. These responses were, however, largely non-neutralizing and displayed a capacity to enhance dengue infection in vitro at significantly low dilution (1:10). In contrast, a subject who had high levels of neutralizing antibodies against both dengue and Zika viruses enhanced infection at a dilution of 1:10 000. These results suggest that high levels of dengue cross-neutralizin… Show more

“…On the other hand, observations from arbovirus surveillance in Brazil suggest a decrease in DENV circulation after the ZIKV outbreak, possibly due to DENV cross-neutralization by ZIKV antibodies [80]. Additionally, there are indications that these cross-neutralizing ZIKV antibodies can prevent DENV ADE [81, 82]. However, for DENV, it is demonstrated that the risk of severe disease depends on the titer of preexisting DENV antibodies [16].…”

“…However, if this threshold is not reached, neutralization fails, and infection might be enhanced. An in-depth clinical study revealed a history of vaccination against Japanese encephalitis virus two years earlier, and thus, in vitro neutralization could be mediated by antibodies binding to common epitopes among flaviviruses [43]. The use of Mexican samples was valuable because this population is from highly endemic areas of DENV; furthermore, they might have been infected by more than one DENV serotype since more than one serotype could be circulating at the same time or cyclically.…”

“…Recently, several studies have indicated that pre-existing DENV immunity mediated cross-protection against secondary ZIKV infection in mice (Pantoja et al, 2017; Wen et al, 2017). Valiant et al demonstrated that high levels of cross-neutralizing antibodies (nAbs) against DENV in ZIKV seropositive individuals could potentially reduce the enhancement of DENV infection (Valiant et al, 2018). However, some inconsistent results on the outcome of cross-reactive immune responses between DENV and ZIKV have also been reported.…”
