Human Macula Investigated In Vivo with Polarization-Sensitive Optical Coherence Tomography

Abstract: PS-OCT represents a powerful tool for increasing image contrast in ocular tissues. The observed polarization-scrambling nature of the RPE may be used in diseased eyes to locate the RPE or remains of the RPE definitively in OCT images.

“…For example, loss of RNFL birefringence is observed during early stages of glaucoma, a leading cause of blindness in the developed world [8,9]. PS-OCT has found many applications, including anterior [10][11][12][13] and posterior eye imaging [13][14][15][16], skin imaging [17][18][19][20], burn depth and thermal damage assessment [21][22][23][24], dental imaging [25,26] and atherosclerotic plaque characterization [27][28][29].…”

“…Polarization-sensitive OCT (PS-OCT), which is one of the functional extensions of OCT, can improve the image contrast of OCT by imaging birefringence of biological fibrous tissues [4]. PS-OCT has been demonstrated to discriminate between ocular tissues using their birefringence properties in studies on the posterior segment of the eye, retinal nerve fiber [5,6] and scar tissue in age-related macular degeneration [7,8]. Simultaneous polarization scrambling measurements discriminated the retinal pigment epithelium from other structures [7,9].…”

Simultaneous polarization scrambling measurements discriminated the retinal pigment epithelium from other structures [7,9]. In the anterior eye segment, birefringence of trabecular meshwork and sclera has been used to discriminate individual tissues [10].…”

“…Mueller matrix methods are often used to reconstruct polarization parameters [42,43]. Depending on generator and analyzer configuration, PS-OCT can be used for simultaneous depthresolved determination of reflectivity, retardation, slow axis orientation, and relative attenuation [44][45][46][47][48]. Problems with this approach include speckle noise, inaccuracy in refractive index estimates, and high cost.…”
