Human Hormone Production in vitro

Abstract: A new in vitro hormone-synthesizing cell system, line BeWO, derived from human gestational choriocarcinoma, has been permanently established; a high degree of synthesis of functional hormone has been maintained for 18 months in tissue culture. Predominantly glycolytic metabolism, dependent on the level of glucose in the medium, has been observed. Many facets of placental and tumor-cell metabolism, growth, and differentiation are found in this unique multipotential cell.

“…Thus, in most cases, choriocarcinoma has the appearance of trophoblast, being predominantly syncytiotrophoblastic or cytotrophoblastic. Some cytotrophoblastic choriocarcinomas secrete little human chorionic gonadotropin and some choriocarcinomas also secrete human placental lactogen [22][23][24][25][26][27]. A number of choriocarcinoma cell lines have been established; these replicating trophoblasts, derived from the malignant tumor or produced by viral transformation of normal trophoblasts, are appropriate systems to mimick trophoblast behaviour in vitro.…”

“…BeWo, JAr, and Jeg3. The BeWo cell line which is heterogenous on several criteria [22], is comprised of cytotrophoblasts with no differentiation to syncytium [23] under non activated conditions [24]. This is in contrast to primary cultures of term cytotrophoblasts which aggregate and form syncytia.…”

“…Among these are naturally immortalized cell lines obtained from choriocarcinoma tissue, such as BeWo [16], JAR [17] and JEG-3 [18]. The cell lines AC1M-32 [19] and ACH-3P [20] have been generated by fusion of the AC1-1 cell line (a JEG-3 mutant [21]) with primary term or first trimester trophoblasts, respectively.…”

“…BeWo cells are derived from a malignant choriocarcinoma (23), and thus may have a number of differences when compared to normal cytotrophoblasts and syncytiotrophoblasts. Therefore, we used primary cultures of trophoblasts, obtained from placentas harvested after normal-term pregnancies, to examine E-cadherin expression and the role of InlA in bacterial attachment and entry.…”
