Human growth hormone in cultures of human pituitary tumors

Abstract: ✓ Tissue culture medium of 14 human pituitary adenomas removed from acromegalic and hypopituitary patients was analyzed for human growth hormone (HGH). High HGH levels were detected in the culture medium of all adenomas removed from acromegalic patients. HGH was undetectable in the tissue culture medium of six of eight adenomas removed from hypopituitary patients. Elevated HGH levels were also found in the culture medium of pituitary tissue obtained from a juvenile diabetic patient.

“…In addition, there were small clusters or rosettes of epithelioid cells which could be easily dislodged from the surface of the culture flask. Similar findings have been noted in the culture media of human pituitary fragments by other investigators (Batzdorf et al 1971;Teraoka 1972;Tixier-Vidal 1975) and in cultures of other endocrine tissues such as the parathyroid and the thyroid glands (Lissitzky et al 1971). …”

“…Other investigators have attempted to assay hormones in the culture media from chromophobe adenomas with variable results (Kohler et al 1969;Batzdorf et al 1971;Teraoka 1972). Low concentrations of HGH and TSH have been demonstrated.…”

“…However, most other investigators such as Betzdorf et al (1971), Peillon et al (1975) and Kageyama et al (1976) reported the duration of GH secretion to be 2-4 weeks. Minute amounts of GH secretion have been reported by Kageyama et al (1976) in some tumors from nonacromegalic patients, but the amount seemed to be too small to describe to tumoral secretions.…”

“…Köhler et al (1969) obtained hGH values between 50 and 400 ng/day culture and hGH production was sustained for up to one year. Batzdorf et al (1971) found hGH concentration between 0.46 and 13 pg/ml in somatotrophic adenoma culture media maintained for 7 or 14 days in culture. hGH production appears important in our study and high levels are observed in some instances (range 100 to 200 pg/ml).…”
