HPLC‐ESI‐MS/MS analysis and pharmacokinetics of luteoloside, a potential anticarcinogenic component isolated from Lonicera japonica, in beagle dogs

Abstract: Luteoloside is a potential anticarcinogenic component isolated from Lonicera japonica, a traditional Chinese medicine (TCM). This study details the development and validation of a sensitive and accurate HPLC-ESI-MS/MS method for the quantification of luteoloside in dog plasma. Sample pretreatment includes simple protein precipitation using methanol-acetonitrile (1:1, v/v). A Phenomenex Gemini C(18) column (2.0 × 50 mm, i.d., 3.5 µm) was used to separate luteoloside and internal standard by gradient mode with m… Show more

“…Flavonoids are well known as ROS scavengers. As luteoloside is a kind of flavonoid isolated from Chinese herb [30], we investigated whether the intracellular ROS is part of the mechanism by which luteoloside suppress the proliferation, migration and invasion potential of HCC cells. We found that luteoloside could significantly decrease the ROS level of Huh-7 and SMMC-7721 cells in a dose-dependent manner (Fig.…”

“…In order to further demonstrate the superiority of our method, we compared our method with previous reported [1,2,6,8,35,36]. As seen from Table 4, in comparison with other methods, MSPE method had higher extraction ability, thereby it illustrated our method not only had better extraction efficiency but also shortened extraction time to a great extent.…”

“…Several methods have been reported for the determination of flavonoids in different matrices, including HPLC-ESI-MS/ MS in traditional Chinese medicine (Lonicera japonica) [6] and in red wine [7]. HPLC-UV in Echinophora platyloba [8], HPLC-UV in plasma [9] and urine [10], HPLC with fluorometric detection in plasma and urine [11], and a potential method [12].…”

“…Researchers thought most of these effects may be related to the active compositions of volatile oil, chlorogenic acid, and flavones. Chlorogenic acid and luteoloside, officially used as the indicator compound to characterize the quality of this herb and its related preparations, were shown to have beneficial effects in the aspects of anti-oxidation and antitumor [1,13]. Moreover, chlorogenic acid showed the antibacterial, antiviral, anti-inflammatory, hypoglycemic activities, and allergy-preventive properties [1,14].…”
