At present, the identification of honeysuckle aroma depends on experienced tasters, which results in inconsistencies due to human error. The key odorants have the potential to distinguish the different species and evaluate the quality of honeysuckle. Hence, in this study, a more scientific approach was applied to distinguish various honeysuckles. The volatile compounds of different species and parts of honeysuckle were separately extracted by headspace-solid phase microextraction (HS-SPME) and solvent assisted flavor evaporation (SAFE). Compounds with greater volatility such as aldehydes, limonene, γ-terpinene, and terpinolene were preferentially extracted by HS-SPME. As a complementary extraction method to HS-SPME, SAFE was found to recover comparatively more polar compounds such as eugenol, decanoic acid, and vanillin. Subsequently, key odorants with the highest flavour dilution (FD) factors were detected by aroma extract dilution analysis (AEDA). These were benzaldehyde, 4-ethylphenol, decanoic acid, vanillin, 3-methyl-2-butenal, and β -ionone in honeysuckle flowers and γ -octalactone, 4-ethyl phenol, and vanillin in honeysuckle stem. Finally, principal component analysis (PCA) was conducted to analyze not only the key odorants of species and parts of honeysuckle but also their different origins. The results of PCA suggested that the species of honeysuckle contributed much more to variations in aroma rather than their origins. In conclusion, the application of the key odorants combined with PCA was demonstrated as a valid approach to differentiate species, origins, and parts of honeysuckle.

Both Lonicerae japonicae flos and Lonicerae similis flos are important components in traditional Chinese medicine (TCM) with precious medicinal value. However, the absence of studies on their chloroplast genomes and chromatography has considerably hindered the study of their evolutionary and phylogenetic relationships. In this study, the complete chloroplast (cp) genomes of Lonicera acuminata Wall. and Lonicera similis Hemsl. were sequenced using the Illumina sequencing platform and compared with that of Lonicera japonica Thunb., which has been previously reported. Furthermore, the chromatographic fingerprints of the three plants were constructed using HPLC and the content of quality marker (Q-Marker) was calculated. The annotation results showed that the two chloroplast genomes were typical quadripartite structures with lengths of 155,330 bp (L. acuminata) and 155,207 bp (L. similis). A total of 126 different genes were annotated, containing 82 protein-coding genes, 36 tRNA genes, and 8 rRNA genes. The expansion and contraction of the inverted repeat (IR) regions suggested that the boundary regions of IR/SC were comparatively conserved in the three species, and six regions (trnH-GUG-psbA, rps2-rpoC2, rbcL-psaI, trnN-GUU-ndhF, rps15-ycf1, and infA) with nucleotide diversity values (Pi) of variable sites higher than 1% were identified. Phylogenetic relation indicated that L. similis had a closer genetic relationship with L. japonica than L. acuminata. Additionally, the chromatographic fingerprints showed that the characteristic peaks of the three medicinal plants were similar, including Neochlorogenic acid, Chlorogenic acid, 4-Dicaffeoylquinic acid, Sweroside, Secoxyloganin, Luteoloside, Isochlorogenic acid A, Isochlorogenic acid B, and Isochlorogenic acid C. The content of chlorogenic acid and total phenolic acid in L. acuminata (7.4633 ± 0.4461%, 14.8953 ± 0.0728%) and L. similis (14.1055 ± 0.2566%, 21.9782 ± 0.1331%) was much higher than that of L. japonica (3.9729 ± 0.0928%, 6.0964 ± 0.1228%), respectively. This study provides appropriate information for species identification, phylogeny, quality assessment, and rational use of three medicinal plants of the genus Lonicera.

Standardization of Lonicera Japonica Flowering Bud and Contents of Chlorogenic, Rosmarinic and Caffeic Acids in Selected Thai Medicinal Plants

Quantification of chlorogenic, rosmarinic and caffeic acids in 111 selected Thai medicinal plants using high performance liquid chromatography demonstrated that among 111 samples, 39.64% contained all of 3 compounds, 40.54% contained 2 compounds, 14.41% contained only 1 compound and 5.41% could not detect these 3 compounds. Lonicera japonica flowering buds were found to be the richest source for chlorogenic acid content, Melissa officinalis leaves showed the most rosmarinic acid content and the most caffeic acid content was found in Coffea canephora seeds. Pharmacognostic specification and chlorogenic acid content of L. japonica flowering bud from 15 various herbal drugstores throughout Thailand were established. Macroscopic and microscopic evaluation of flowering bud were demonstrated. Physico-chemical parameters including loss on drying, total ash, acid insoluble ash, water content, ethanol and water soluble extractive values were found to be 10.11, 6.59, 1.14, 10.82, 16.46 and 28.88 % by dry weight respectively. For quantitative analysis, chlorogenic acid content in flowering bud by TLC-densitometry compared to TLC-image analysis by imageJ software were found to be 2.24 and 2.09 g/100 g respectively which were not significantly different (P = 0.13). The validation parameters of all quantitative analysis were investigated according to ICH guideline. HPLC as well as TLC-densitometry and TLC-image analysis were demonstrated as suitable, reliable and efficient methods for the quantitative analyses. In vitro biological activities of L. japonica flowering bud compared to chlorogenic, rosmarinic and caffeic acids were evaluated by brine shrimp lethality assay, MTT cell viability assay, comet assay, antimicrobial activities, antioxidant activities and yeast alpha-glucosidase inhibition assay. The results demonstrated that flowering bud ethanolic extract showed non-toxicity on brine shrimp nauplii and 6 tested cell lines. Chlorogenic, rosmarinic and caffeic acids demonstrated toxicity against brine shrimp nauplii. They showed more cytotoxic potentials against tested cell lines than the extract but were still accepted as no cytotoxicity. The extract and 3 compounds showed human lymphocyte DNA damage by comet assay. They were no inhibitory activities against tested microorganisms. The extract and the compounds demonstrated the abilities of DPPH and nitric oxide scavenger and reducing power. However, only the compounds exhibited beta-carotene bleaching activity. Moreover, they inhibited enzyme activity in yeast alpha-glucosidase inhibition study.
