How to: Measuring blood cytokines in biological psychiatry using commercially available multiplex immunoassays

Abstract: Cytokines produced by both immune and non-immune cells are likely to play roles in the development and/or progression of psychiatric disorders. Indeed, many investigators have compared the blood cytokine levels in psychiatric patients with those of healthy controls or monitored their levels in patients during disease progression to identify biomarkers. Nevertheless, very few studies have confirmed that such cytokines remain stable in healthy individuals through periods of weeks and months. This is an important… Show more

“…We sought to replicate the serum biomarker proteins identified on the RBM platform by subsequently analyzing proteins on a Meso Scales Discovery (MSD) platform [19] run in-house. Compared with the traditional ELISA approach, the MSD platform shows greater sensitivity and is able to reliably detect different proteins across a broad dynamic range of concentrations [20].…”

“…The BALF and cell culture supernatant levels of IL-1β, IL-6, IL-10, and TNF-α were analyzed using the Bio-Plex Pro Mouse Cytokine 6-plex Assay (Bio-Rad, product No. M6000007NY) according to the manufacturer's instructions in the Bio-Plex 200 system (Bio-Rad, Hercules, CA) as previously reported (15).…”

“…Furthermore, PDGF-BB was found to be increased in BD patients suffering a depressive episode after treatment with a combination of sleep deprivation, lithium and bright light therapy (29). PDGF-BB was also found to have low intraindividual variability when measured with different methods and in serum and plasma, suggesting that this marker may be technically reliable (30). Physiologically, PDGF receptors have been reported to play a role in glutamatergic signaling (31), which is thought to be dysregulated in subtypes of affective patients (32).…”
