How Do Ethanolamine Plasmalogens Contribute to Order and Structure of Neurological Membranes?

Abstract: Ethanolamine plasmalogen (EtnPLA) is a conicalshaped ether lipid and an essential component of neurological membranes. Low stability against oxidation limits its study in experiments. The concentration of EtnPLA in the bilayer varies depending on cell type and disease progression. Here we report on mixed bilayers of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and 1-(1Z-octadecenyl)-2-oleoyl-sn-glycero-3-phosphoethanolamine (C18(Plasm)-18:1PE, PLAPE), an EtnPLA lipid subtype, at mole ratios of 2:1, … Show more

“…PC and PA or PG are randomly placed in a 3:1 molar ratio, PC and CL are in a 6:1 molar ratio, but one anionic lipid was placed near each of intermonomer groove. The CHARMM36m parameters were used for protein and lipids, TIP3P was for water (48, 49). The parameters for ions were taken from the previous report (50).…”

“…The number of lipid molecules (128 total) was chosen based on recent reports. [65][66][67] Starting coordinates of the lipid bilayer, composed of 128 molecules of DMPC, were taken from the website of Biocomputing laboratory at the University of Calgary (http:// wcm.ucalgary.ca/tieleman/downloads) and solvated with 5227 water molecules in a rectangular box of 6.5 × 6.5 × 7.0 nm 3 . The mixed bilayers of DMPC and gemini components were obtained by a random selection of 52 molecules of DMPC replaced by the same number of gemini molecules.…”

“…However, with plasmalogen deficiency, the total amount of PUFAs in phosphatidylethanolamine remains constant in human fibroblasts and in the brains of mice. Plasmalogens have been noted to play an important role as neuroprotectors and modulators of the signaling mechanisms of cell membranes [ 85 ]. Plasmalogens also act as endogenous antioxidants, protecting lipids and lipoproteins from oxidative stress [ 86 ].…”
