Hormonal regulation of interleukin-6 production in human adipocytes

Abstract: OBJECTIVE:To elucidate the hormonal regulation of interleukin-6 (IL-6) production by human adipose tissue and its relation to leptin. DESIGN: In vitro study. Human adipocytes were incubated with dexamethasone (with or without RU486), norepinephrine and epinephrine (with or without propranolol), or insulin. MEASUREMENTS: IL-6 and leptin secretion by human adipocytes. RESULTS: A gradual increase in IL-6 secretion by adipocytes during differentiation was observed. A positive correlation was found between basal IL… Show more

“…In accordance with these findings, IL-6 protein secretion was increased in human adipocytes in vitro and in vivo in volunteers after treatment with the β-adrenoceptor agonist isoprenaline [47,48,49]. Furthermore, insulin stimulates and glucocorticoids suppress IL-6 mRNA and protein expression in 3T3-L1 and human adipocytes (Table 1) [46,48,49,50]. In accordance with these in vitro results, diminished exercise-induced IL-6 plasma concentrations have been found after treatment with hydrocortisone or dexamethasone in vivo [51].…”

“…β-adrenergic activation stimulates IL-6 mRNA expression in 3T3-L1 adipocytes and this effect is mediated via a classic G S -protein-coupled pathway (Table 1) [46]. In accordance with these findings, IL-6 protein secretion was increased in human adipocytes in vitro and in vivo in volunteers after treatment with the β-adrenoceptor agonist isoprenaline [47,48,49]. “…In addition, the sympathetic nervous system may be an important regulator of the synthesis and release of IL-6 from adipocytes. 8 IL-6 release from cultured human adipocytes can be inhibited by glucocorticoids and induced by insulin, 9 and a short-term b-adrenergic stimulation elevates IL-6 concentrations by increased release from fat depots and muscle. 10 The influence of diet and its fatty acid composition on adipose tissue development and distribution is well known, 11,12 as is the close relation between nutrition-and adipose tissue-associated diseases.…”
