HLA Haplotypes and Class II Molecular Alleles in Argentinian Patients with Pemphigus Vulgaris

Abstract: In patients with the haplotype DRb1* 0402/DQb1* 0302 the affectation of 10%-30% of the corporal surface prevailed (ACS). In patients with DRb1* 1401/DRQb1* 0503, involvement of <10% of the ACS prevailed.

“…Our results demonstrated with a high rate of reliability that the presence of the DRB1*04 and DRB1*14 alleles is related to a high risk for PV because almost all of the present patients (99.0%) carried one of these two alleles, whereas only 35.0% of the healthy control group did so. The positive association of PV with these two alleles is broadly spread across races and populations …”

“…To date, the major histocompatibility complex (MHC) seems to represent the only region to be clearly involved in the genetic predisposition to PV . Many human leukocyte antigen class II (HLA‐II) alleles have been considered as risk factors in different populations . However, in Syria, where PV is not uncommon, no studies have as yet shed light on the role of HLA polymorphism in PV occurrence.…”

“…9 The theory that PV is inherited had been initially suggested by the unique susceptibility of some ethnic groups and populations such as Ashkenazi Jewish and people from India and the Mediterranean. 16 The association between HLA class II alleles and PV has been shown in many different ethnic groups, such as Brazilians, 13 Venezuelans, 3 Argentinians, 17 Spanish, 18 French, 19 Iranians, 20 Italians and Sardinians, [21][22][23] Japanese, [24][25][26][27] Ashkenazi Jews, 28 Mexicans, 29 North-Americans, [30][31][32] Pakistanis, 33 Syrians, 10 Middle-Eastern groups, 16 Slovacs, 34 and Canadians, 35 associating nine DR alleles DRB1*0402, *0403, *0406, *0802, *0804, *1401, *1404, *1405, *1408 and four DQ alleles DQB1*0302, DQB1*0503, DQA1*0101, and DQA1*0301 with the disease. 11,12 The association between PV and human leukocyte antigen (HLA) was shown in the 1980s and has repeatedly been proven through laboratory and serologic tests.…”

“…25,26 HLA alleles seem to be the most important predisposing genetic factors. 26,27 As to the association with class I HLA system, Glorio et al did not find an association of PV with HLA-A, -B or -C antigens in Caucasian Argentinians. 27 Miyagawa et al found a high prevalence of HLA-B15 antigen, specifically of HLA-B*1507 allele in the Japanese population with PV, 28 and Abroobaker et al, in South Africa, reported positive HLA-B8 antigen.…”

“…28 In Caucasian Argentinians with PV, Glorio et al reported an increased frequency of HLA-DR3 and -DR4 antigens, of HLA-DRB1*0402, -DQB1*0302, -DRB1*1401 and -DQB1*0503 alleles. 27,31 In Italy, a high frequency of HLA-DRB1*04 and -DRB1*1401 alleles was seen both in individuals with PV and PF, with protection being associated with the HLA-DRB1*07 allele. 11 In Japanese individuals with PV there was a positive association with HLA-DQB1*0503 and -DRB1*1405 alleles and a negative association with HLA-DQA1*0103 and -DQB1*0601 alleles.…”
