HIV-1 and HIV-2 Vif Interact with Human APOBEC3 Proteins Using Completely Different Determinants

Abstract: Human APOBEC3 (A3) restriction factors provide intrinsic immunity against zoonotic transmission of pathogenic viruses. A3D, A3F, A3G, and A3H haplotype II (A3H-hapII) can be packaged into virion infectivity factor (Vif)-deficient HIVs to inhibit viral replication. To overcome these restriction factors, Vif binds to the A3 proteins in viral producer cells to target them for ubiquitination and proteasomal degradation, thus preventing their packaging into assembling virions. Therefore, the Vif-A3 interactions are… Show more

“…We also conducted western blots analysis to probe for APOBEC3C (S188 and I188) expression in the presence of HIV-1 and HIV-2 Vif proteins ( Figure 3.9B). Consistent with other reports, A3C S188 protein levels are significantly decreased in the presence of HIV-1 Vif [149,222] as well as HIV-2 Vif [225]. Likewise, the expression of the A3C I188 variant also dramatically decreased in the presence of HIV-1 and HIV-2 Vif.…”

“…Intriguingly, HIV-2 ∆ vif has been reported to be less sensitive than HIV-1 ∆ vif to APOBEC3G [ 32 ], but susceptibilities to other APOBEC3 proteins (in the presence or absence of Vif) have not yet been compared. Further, HIV-1 and HIV-2 Vif were recently shown to interact with APOBEC3F and APOBEC3G through completely separate sequence determinants, and differences in HIV-1 and HIV-2 Vif-induced degradation of specific APOBEC3 proteins were also noted [ 60 ].…”

“…Nonetheless, as the mRNAs corresponding to the human versions of the APOBEC3 proteins are widely expressed [ 27 ], it is likely that the single viral protein Vif would need to simultaneously adapt to multiple host restriction factors with different interfaces to allow viral replication in chimpanzees. Furthermore, Vif has distinct regions for antagonizing the APOBEC3 proteins [ 19 , 42 , 43 ]. Therefore, overcoming one APOBEC3 protein after adaptation will not necessarily allow the virus to overcome the other APOBEC3 members.…”
