Background: Insecticide-resistant Culex quinquefasciatus mosquitoes have been reported worldwide but the susceptibility status of Cx.quinquefasciatus larvae to insecticides has not been determined in Jakarta, Indonesia. This study aimed to evaluate the susceptibility status of Cx. quinquefasciatus larvae to temephos, malathion, cypermethrin, and deltamethrin in Jakarta, Indonesia, through the mode of action detoxification enzyme activity and digestive system damage.Methods: Cx. quinquefasciatus larvae were collected from five sites in Jakarta. The larval susceptibility test to temephos used the WHO standard kits (1.25, 6.25, 31.25, and 156.25 ppm) and to malathion (0.5 ppm), cypermethrin (.25 ppm), and deltamethrin (0.35 ppm). The activity of acetylcholinesterase (AChE), glutathione S-transferase (GST), and oxidase were measured by the biochemical method. The larval midgut cell damage was examined using the routine histopathological method and transmission electron microscopy (TEM).Results: At 24 h, temephos and deltamethrin caused 100% larval death of Cx.quinquefasciatus in five sites of Jakarta. Temephos and malathion significantly inhibited AChE activity, while cypermethrin and deltamethrin significantly inhibited oxidase activity. All tested insecticides significantly damaged the larval midgut. The TEM findings showed the damaged parts of the midgut epithelial cells included the cell membrane, nuclei, nucleoli, mitochondria, and other organelles cells.Conclusion: This study is the first to report that Cx.quinquefasciatus larvae in Jakarta were completely susceptible to temephos and deltamethrin through metabolic enzymes and midgut cell damage. This study provides baseline data to improve mosquito control programs.

Nyamuk Aedes aegypti merupakan vektor utama penyakit Demam Berdarah Dengue (DBD). Berbagai upaya pencegahan dilakukan untuk menurunkan populasi nyamuk Aedes aegypti. Salah satunya dengan menggunakan insektisida baik sintetik maupun organik. Bioinsektisida menjadi alternatif yang banyak digunakan saat ini karena lebih aman terhadap manusia, hewan dan lingkungan. Larvasida merupakan salah satu bentuk insektisida yang menyerang tempat perindukan nyamuk di air. Larvasida bekerja dengan berbagai mekanisme dan menyerang berbagai target organ termasuk midgut larva. Berdasarkan beberapa penelitian, insektisida nabati terbukti menyebabkan sel epitel midgut larva menjadi lisis, berubah bentuk, vakuolisasi sitoplasma dan terlepas dari membran basal. Membran peritrofik, mikrovili dan brush border mengalami kerusakan parsial maupun total. Perubahan histopatologis sel epitel midgut larva Aedes aegypti ini disebabkan oleh senyawa metabolit baik tunggal maupun kombinasi yang terdapat dalam ekstrak tanaman.

Filariasis, transmitted by the lymphatic filariasis vector Culex quinquefasciatus, is still a global public health issue. Resistance to Cx. quinquefasciatus is caused by the eradication of filariasis with synthetic insecticides. The aim of the study was to evaluate the toxicity of pure camphor on Cx. quinquefasciatus, with a focus on detoxifying enzymes and midgut histopathological abnormalities. Cx. quinquefasciatus larvae are field-collected wild-strain larvae. The WHO protocol was followed for the larval bioassays. Pure camphor concentrations of 0.5, 1.5, 10.5, 25.5, and 50 ppm were applied to larvae for 24, 48, and 72 h, with 5 replications. The biochemical method was used to test detoxification enzymes such as acetylcholinesterase (AChE), glutathione S-transferase (GST), and oxidase. The routine histopathological examination was performed on the larvae's histopathological midguts. At 48 h, pure camphor caused 100% mortality of Cx. quinquefasciatus larvae. LC50 and LC90 values were found to be 2.32 and 12.40 ppm respectively. AChE and oxidase activity were both significantly lower (p<0.05). Damage to the food bolus (FB) and peritrophic membrane (PM ), broken epithelial layers (EP), changes in the size and shape of epithelial cells (EC), and microvilli (M v) damage were all observed in larval midguts. In our study, pure camphor demonstrated larvicidal activity via decreased AChE and oxidase act ivity, as well as midgut damage.
