Histocompatibility Phenotyping by the Mixed Lymphocyte Reaction

Abstract: A study of seven pig families enabled the identification of individuals who were homozygous for the gene products identified by the mixed lymphocyte reaction. Homozygotes were engaged as markers to elucidate histocompatibility phenotypes. Thus a system evolved in which histocompatibility phenotypes were assessed independently of serologically dependent techniques.

“…Liver cells, however, stimulated well although they did not express la-like antigens. The conclusion that MHC-encoded LD-Ags on kidney or liver cells are the same as those on lymphocytes suggests that in pigs the single MHC locus coding for lymphocyte LD-Ags (Bradley et al 1973) is separate from the locus coding for la-like antigens.…”

“…Large White pigs were obtained from the herd at the Agricultural Research Council Institute at Babraham. Lymphocyte separation and culture techniques were as described previously by Bradley et al (1972Bradley et al ( , 1973a. Lymphocytes from each individual were divided into two aliquots, one of which was irradiated to prevent mitosis.…”

“…In previous work outbred pigs which were homozygous for gene products causing stimulation in the mixed lymphocyte reaction ( M L R ) were demonstrated (Bradley et al 1972). These were employed as genetic markers to phenotype families (Bradley et al 1973a). Reactions to homozygous stimulator cells were either zero or positive, according to whether stimulator gene products were present or absent i n responder cells.…”

“…Because the heart transplantation was decided by the doctors, the child was submitted on several examinations to check his eligibility. One of the first examinations required was measuring the histocompatibility of Lymphocyte Reaction [7,8]. This test showed almost 100% reaction of antibodies in this patient, and the acceptable level is under 10% for the transplantation to take place at this institution.…”
