Histamine release test in the diagnosis of human hydatidosis

Abstract: An evaluation of the use of the histamine release test (HRT) in the diagnosis of human hydatidosis is presented. This technique, which makes it possible to evaluate directly IgE dependent immediate hypersensitivity by measuring the histamine released from leucocytes (basophils) after antigenic challenge, was compared with the detection of serum specific IgE by the radio-allergosorbent test (RAST), and with the determination of serum specific IgE by the enzyme-linked immunosorbent assay (ELISA). Of the 54 patie… Show more

“…Wc have reported that in basophils from hydatid pa tients the antigen-induced histamine release correlates neither with serum nor with cell-bound specific IgE [8], From the present study it is apparent that E. granulosus in fection induces both generation of histamine releasing factor and production of IgE that can bind this cytokine. Thus, it is possible that these two events can influence the response of basophils to antigenic challenge.…”

“…Indeed, this hypcrrcleasable state makes it possible to obtain a high response to appropriate antigen, even though levels of cell-bound IgE are low [8].…”

“…basophil) depends also on its 'releasability', that is the intrinsic capacity of the cell to release mediators in response to a variety of immunologic and non-immunologic stimuli [1], We have studied the antigen-specific histamine release from blood basophils in subjects with hydatidosis [2,3]. In our patients, a high rate of histamine liberation was noted in all cases, even though the levels of serum reaginic antibodies were undetectable.…”

“…Histamine release experiments were carried out following the procedure previously described [3]. Histamine content was as sessed by the automated fluorometric assay [4], The following stimuli were used for challenge: calcium ionophore A23187 (Calbiochem); formyl-methionylleucyl-phenylalanine (f-met peptide; Sigma); goat anti-human IgE (Sigma), and hydatid antigen, pre pared according to the method previously described [3]. IgE was dissociated from basophils of 12 patients with hydatidosis by acetate elution [5].…”
